【摘要】 目的 :研究中华灵芝宝的抗肿瘤作用机理。方法 :DNA超螺旋解旋法检测中华灵芝宝对拓扑异构酶Ⅰ、Ⅱ (TOPOⅠ、Ⅱ )活性的影响 ;琼脂糖凝胶电泳检测其对DNA的直接打断作用及诱导K5 6 2细胞凋亡的能力。结果 :中华灵芝宝能抑制TOPOⅠ、Ⅱ的活性 ,促进DNA的解旋、断裂 ;同时还能直接打断质粒和大分子DNA ;作用于K5 6 2细胞后电泳显示有凋亡细胞特有的“梯状”条带。结论 :中华灵芝宝能同时抑制TOPOⅠ、Ⅱ活性 ,直接打断质粒和大分子DNA ,并能诱导K5 6 2细胞凋亡。更多还原

【Abstract】 Objective:To study the anticancer mechanisms of China ganoderma lucid essence (CGLE). Methods:The effect of CGLE on topoisomerases was measured by supercoiled DNA relaxation assay;the effects of CGLE on direct breaking of DNA and inducing apoptosis ability of K562 cells were measured by agarose gel electrophoresis. Results:CGLE could markedly inhibit the activities of topoisomerase Ⅰand Ⅱ,promote the relaxing and breaking of DNA.Simultaneously dose dependent breaks of plasmid DNA and nuclei DNA were observed.Besides it could induce apoptosis of K562 cells. Conclusions:These results suggest that CGLE can inhibit the activities of topoisomerase Ⅰand Ⅱ,and directly break plasmid DNA and nuclei DNA.It can also induce the apoptosis of K562 cells.更多还原
