【摘要】 以家兔的基因组ＤＮＡ 为模板，研究影响家兔ＲＡＰＤ 扩增的因素。实验结果表明，Ｔａｑ酶、随机引物和Ｍｇ２ ＋ 浓度及模板ＤＮＡ 的纯度对ＲＡＰＤ 扩增影响较大。本实验建立了适于家兔ＲＡＰＤ 分析的反应体系：反应体积为２０μｌ，其中含有１ ×扩增缓冲液，２ ．０ｍ ｍｏｌ／Ｌ ＭｇＣｌ２ ，０ ．１ｍ ｍｏｌ／ＬｄＮＴＰ，０－２μｍｏｌ／Ｌ 随机引物，１ ．５ＵＴａｑ 酶，９０ｎｇ 模板ＤＮＡ更多还原

【Abstract】 The study on factors affecting amplification of RAPD for rabbit was carried out by using rabbit genomic DNA as template. The result showed that the concentrations of Taq polymerase, random primer, Mg 2+ and the purity of template DNA played an important role in RAPD analysis.A technical system for RAPD analysis in rabbit was established: in 20μl reaction solution, containing 1×buffer, 2 0mmol/L MgCl 2, 0.2μmol/L primer, 0.1mmol/L dNTP, 90ng template DNA, 1 5UTaq polymerase.更多还原
