乙型肝炎病毒对精子染色体的影响
THE EFFECT OF HEPATITIS B VIRUS ON HUMAN SPERM CHROMOSOME
【摘要】 乙型肝炎病毒感染者的精子与去透明带金黄地鼠卵受精制备染色体,并以HBV 全长DNA 作探针对精子染色体进行荧光原位杂交。结果发现慢性迁延型肝炎患者、慢性活动型肝炎患者和HBV 携带者的精子染色体畸变率分别为12-33 % (9/73) 、16-18% (11/68) 和15-48 %(13/48),均显著高于健康对照者精子畸变率(4 .35% ,5/115) 。进一步FISH 显示,在1 例慢性迁延型肝炎患者的精子染色体上有特异荧光信号。研究结果表明,HBV 能导致精子染色体不稳定性,并在患者精子染色体上整合,其整合特点是:整合位点是随机的,且大多为多位点整合。
【Abstract】 Sperm chromosome of subjects with hepatitis B virus infection were prepared using interspecific in vitro fertilization with zona free hamster eggs. Fluorescence in situ hybridization(FISH) to sperm chromosome spreads was carried out with biotin labelled full length HBV DNA. There was significant difference in the frequencies of sperms with chromosome aberrations between subjects with HBV infection and control. FISH showed that HBV DNA integrated into sperm chromosomes, and that multisite and nonspecific integrations were the features of HBV in sperm chromosomes. These results indicate that HBV infection affects on the sperm chromosome broadly.
