采用振荡-静置循环培养策略制备的灵芝菌丝体水提物的抗氧化活性

【摘要】 采用振荡-静置循环培养策略(振荡2.8 d-静置7.3 d-振荡0.2 d-静置0.3 d),在250 mL摇瓶,3、5、50 L发酵罐制备灵芝(Ganoderma lucidum)菌丝体,测定菌丝体生物量、总三萜含量以及菌丝体水提物总三萜、总酚、总糖含量,研究菌丝体水提物对ABTS、DPPH、超氧阴离子和羟基自由基清除率。结果表明:不同发酵模式优化组菌丝体总三萜含量均高于对照组。优化组之间比较,50L发酵罐生物量最高,250mL摇瓶菌丝体总三萜含量最高;3L发酵罐菌丝体水提物总三萜、总糖含量最高,250mL摇瓶、3L发酵罐菌丝体水提物总酚含量较高;3L发酵罐菌丝体水提物对ABTS自由基清除效果最好,IC50值为(12.31±0.23) mg.mL-1;250mL摇瓶菌丝体水提物对DPPH自由基的清除效果最好,IC50值为(4.53±0.24)mg·mL-1;3、50L发酵罐菌丝体水提物对超氧阴离子自由基的清除效果较好,IC50值分别为(12.29±0.61)、(12.66±0.68)mg·mL-1;250mL摇瓶,5、50L发酵罐菌丝体水提物对羟基自由基清除效果较好,IC50值分别为(12.52±0.73)、(11.87±0.46)、(11.57±0.33) mg·mL-1。研究结果可为灵芝液态发酵的工业化生产和应用提供参考。
【Abstract】 Ganoderma lucidum mycelia were cultivated using an oscillation-static cyclic culture strategy(2.8 d oscillation-7.3 d static-0.2 d oscillation-0.3 d static) in 250 mL shake flasks,3 L,5 L,and 50 L fermenters.Mycelial biomass,total triterpenoid content,and the levels of total triterpenoids,total phenols,and total polysaccharides in mycelial aqueous extracts were analyzed.The antioxidant activity of aqueous extracts was evaluated by measuring their scavenging effects on ABTS,DPPH,superoxide anion,and hydroxyl radicals.Results demonstrated that all optimization groups exhibited higher total triterpenoid content in mycelia compared to the control group.Among the optimized conditions:the 50 L fermenter achieved the highest biomass,while the 250 mL shake flask produced mycelia with the highest total triterpenoid content.Aqueous extracts from the 3 L fermenter contained the highest levels of total triterpenoids and polysaccharides,whereas the 250 mL shake flask and 3 L fermenter extracts showed elevated total phenol content.In radical scavenging rate,the 3 L fermenter extract displayed the strongest ABTS radical scavenging(IC50:12.31 ± 0.23 mg·mL-1),while the 250 mL shake flask extract exhibited optimal DPPH radical scavenging(IC50:4.53 ± 0.24 mg·mL-1). For superoxide anion radicals,the 3 L and 50 L fermenter extracts performed best(IC50:12.29 ± 0.61 and 12.66 ± 0.68 mg·mL-1, respectively),whereas hydroxyl radical scavenging was most effective with the 250 mL shake flask,5 L,and 50 L fermenter extracts(IC50:12.52 ± 0.73,11.87 ± 0.46,and 11.57 ± 0.33 mg·mL-1,respectively).These findings provide critical insights for scaling up the industrial production and functional applications of G.lucidum through liquid fermentation.
