一种来自六妹羊肚菌的新型葡聚糖的纯化、结构表征和免疫调节活性研究

【摘要】 为得到具有免疫调节作用的安全的天然化合物。本研究采用水提醇沉结合柱层析的方法从六妹羊肚菌中提取纯化了一种分子量为1.2 × 10~7Da的新型葡聚糖,并命名为MSP-1-1,并通过HPAEC,SEC-MALLS-RI,FTIR,GC-MS,NMR,SEM对其结构进行了表征。结果表明,葡聚糖主链为→4)-α-D-Glcp-(1→连接少量的→4,6) - α - D - Glcp - (1→,支链为α - D - Glcp - (1→连接在糖残基的 0 -6 位→4,6) - α - D - Glcp - (1→。随后,我们对MSP-1-1在体内的免疫应答进行了研究,结果表明MSP-1-1可以恢复环磷酰胺引起的免疫器官指数和外周血水平异常。此外,MSP-1-1通过显著提高巨噬细胞的吞噬功能和脾脏淋巴细胞的增殖功能,上调CD3+CD4+和CD3+CD8+T细胞的比例,提高CD3+CD4+/CD3+CD8+比值,并显著升高血清和胸腺中IgA和IgM的水平,从而发挥免疫调节活性。综上所述,本研究有效地阐明了 MSP-1-1的结构和免疫调节功能,为揭示羊肚菌葡聚糖免疫调节活性的分子机制提供了理论基础。同时,为羊肚菌有价值的功能性产品的开发提供了重要的理论依据。

【Abstract】 In search of natural and safe compounds with immunomodulatory effects.In this research,a novel glucan with molecular weight(Mw) of 1.2×10~7 Da,named MSP-1-1,was extracted and purified from Morchella Sextelata by water extraction and alcohol precipitation combined with column chromatography,and the structure of MSP-1-1 was characterized by HPAEC,SEC-MALLS-RI,FT-IR,GC-MS,NMR,SEM.The findings demonstrated that the glucan backbone was→4)-α-D-Glcp-(1→linked with a small amount of→4,6)-α-D-Glcp-(1→,and the branched chain was α-D-Glcp-(1→ linked at the O-6 position of sugar residue→4,6)-α-D-Glcp-(1→.Subsequently,the immune responses of MSP-1-1 in vivo were investigated,results suggested that MSP-1-1 could restore the immune organ index and peripheral blood level abnormalities caused by cyclophosphamide.In addition,MSP-1-1 significantly improved phagocytosis of macrophages and the proliferation ability of splenic lymphocytes,upregulate the proportion of CD3+CD4+and CD3+CD8+T cells,increase the CD3+CD4+/CD3+CD8+ratio,and significantly elevated the levels of IgA and IgM in serum and thymus to exert immune regulatory activity.In summary,this research effectively elucidated the structure and immunoregulatory function of MSP-1-1,providing a theoretical basis for uncovering the molecular mechanism behind the immuno-regulatory activity of Morchella Sextelata glucan.Moreover,it offers a crucial theoretical basis for the development of valuable functional products of Morchella Sextelata.
