响应面优化灰树花ACE抑制肽的制备工艺

【摘要】 灰树花富含蛋白质,是食源性生物活性肽的良好来源。本研究以灰树花为原料,利用响应面法建立碱性蛋白酶酶解法制备灰树花ACE抑制肽的工艺参数,并对其体外活性进行评价。以ACE抑制率为指标,选取加酶量、酶解温度、酶解时间和料液比为单因素,在单因素实验的基础上,应用Box-Benhnken中心组合实验设计原理进行四因素三水平实验设计,确定碱性蛋白酶酶解灰树花的加酶量、酶解温度、酶解时间等工艺参数。结果表明,碱性蛋白酶酶解制备灰树花源ACE抑制肽的最佳酶解条件为加酶量为1.03%,酶解温度为47℃、酶解时间为2 h,料液比为1:15 g/mL。在此条件下制备的灰树花ACE抑制肽的ACE抑制率为(87.80±0.12)%,与预测值89.413%结果相近,表明该回归模型对优化灰树花酶解制备灰树花源ACE肽工艺可行。
【Abstract】 Grifola frondosa is rich in protein and is a good source of foodborne bioactive peptides.In this study,we used Grifola frondosa as raw material to establish the technological parameters of preparing ACE inhibitory peptide from Grifola frondosa by alkaline protease hydrolysis using response surface method,and evaluated its in vitro activity.The ACE inhibition rate was used as the index,and the enzyme amount,enzyme temperature,enzyme time and solid-liquid ratio were selected as the single factor.On the basis of the single factor experiment,the four-factor and three-level experimental design was carried out by using the Box-Benhnken center combination experimental design principle to determine the enzyme amount,enzyme temperature,enzyme time and other technological parameters of alkaline protease enzymatic hydrolysis of Grifola grifola.The results showed that the optimal conditions for the preparation of ACE inhibitory peptide from Grifola grifola by alkaline protease were as following:enzyme amount 1.03%,enzyme temperature 47℃,enzyme time 2 h,solid-liquid ratio 1:15 g/mL.The ACE inhibition rate of ACE inhibitor peptide prepared under these conditions was(87.80±0.12) %,which was close to the predicted value of 89.413%,indicating that the regression model was feasible for optimizing the enzymatic preparation process of Grifola frondosa ACE peptide.
