3种动物源胶原蛋白肽抗氧化活性研究

【摘要】 在动物产品加工过程中产生的大量副产物,如猪皮、牛皮、鱼鳞和骨骼等,通常会被丢弃。然而,这些副产物是生产高质量胶原蛋白的优质原料。本试验以牛皮、猪皮和鱼鳞为原料,分别使用中性和碱性蛋白酶、风味蛋白酶、胰蛋白酶和木瓜蛋白酶,在其最适pH和温度条件下进行酶解,检测其酶解产物的水解度、蛋白回收率及分子量分布,同时探究其抗氧化活性。结果显示,碱性蛋白酶和胰蛋白酶酶解产物的水解效果和蛋白回收效果较佳。分子质量分布表明,碱性蛋白酶和木瓜蛋白酶更易生成小分子肽。对以上15种酶解产物的DPPH自由基清除能力、羟基自自基清除能力及Fe2?螯合能力进行了测定,结果显示,中性蛋白酶酶解产物的DPPH自由基清除能力较强,3种酶解产物的清除率分别为(5.89±0.88)%,(3.23±0.67)%,(7.03±0.98)%;碱性蛋白酶酶解产物在羟基自由基清除能力方面表现出色,清除率分别为(78.54±4.82) %,(85.06±4.98)%,(73.16 ± 3.95)%;而碱性蛋白酶酶解产物在Fe2?螯合能力方面表现最优,清除率分别为(99.09±0.10)%,(86.97±0.94) %,(97.39±0.10)%。

【Abstract】 A substantial amount of by-products,such as pigskin,bovine hide,fish scales,and bones,are generated during animal product processing and are often discarded.However,these by-products serve as excellent raw materials for producing high-quality collagen.In this study,bovine hide,pigskin,and fish scales were enzymatically hydrolyzed using neutral protease,alkaline protease,flavor protease,trypsin,and papain under their optimal pH and temperature conditions.The degree of hydrolysis,protein recovery rate,and molecular weight distribution of the hydrolysates were evaluated,alongside an investigation into their antioxidant activities.Comparative analysis revealed that hydrolysates from alkaline protease and trypsin exhibited superior hydrolysis efficiency and protein recovery.Molecular weight distribution indicated that alkaline protease and papain more effectively generated low molecular weight peptides.Additionally,the DPPH radical scavenging activity,hydroxyl radical scavenging activity,and Fe~2? chelating ability of the fifteen hydrolysates were measured.Results showed that neutral protease hydrolysates had strong DPPH radical scavenging activity,with scavenging rates of(5.89±0.88) %,(3.23±0.67) %,and(7.03±0.98) %.Alkaline protease hydrolysates demonstrated excellent hydroxyl radical scavenging activity,with rates of(78.54±4.82) %,(85.06±4.98) %,and(73.16±3.95) %,and exhibited the highest Fe~2? chelating ability,with rates of(99.09±0.10) %,(86.97±0.94) %,and(97.39±0.10) %.
