不同生产工艺的次氯酸溶液在模拟临床条件下稳定性的研究
An experimental study on the stability of hypochlorous acid solution produced from different techniques under clinical conditions

【摘要】 目的:探索两种生产工艺制备的次氯酸溶液在模拟临床使用及储存条件下其理化性能的变化,并研究了溶液有效氯浓度的改变对粪肠球菌杀菌效果的影响。方法:准备化学法与电解法两种生产工艺的次氯酸溶液样品((1)化学法制备100 ppm,(2)电解法制备100 ppm,(3)化学法制备200 ppm,(4)电解法制备200 ppm),在模拟临床使用条件下(20-25℃下避光封盖保存,每日开盖5分钟,倒置摇晃2次)监测各溶液样本在1个月内其p H值、氧化还原电位值(oxidation-reduction potential,ORP)、有效氯浓度(free available chlorine,FAC)的变化;并通过悬液定量杀菌试验检测次氯酸溶液随FAC的衰减其对粪肠球菌(E.faecalis)杀菌对数值的改变。另外,将上述4组溶液样品分别开封或未开封保存于室温、避光处,分别于第6个月、第12个月时检测FAC,获得各样品FAC损失率。结果:在模拟临床使用的条件下,各次氯酸溶液的pH值及氧化还原电位值相对稳定,FAC则随时间的延长而缓慢衰减,1个月内衰减程度均<20 ppm;样品(1)-(4)的FAC损失率分别为14.05%、19.55%、7.77%和9.18%,化学法制备的次氯酸溶液其FAC衰减率小于电解法制备的(p<0.05)。两种常规浓度(100 ppm、200 ppm)的次氯酸溶液FAC降低约20 ppm后,其对E.faecalis的杀菌对数值仍可>5.00,满足99.999%的悬液杀菌效果。另外,比较4组样品溶液在开封与未开封状态下的长期FAC稳定性,第12个月时未开封状态下FAC损失率仅为4.0%-17.6%,远低于开封状态(55.0%-99.0%)。结论:两种不同生产工艺的次氯酸溶液在模拟临床使用与储存条件下,其FAC稳定性良好,均可达到较好的杀菌效果;且化学法制备的次氯酸溶液FAC稳定性优于电解法。为次氯酸溶液在根管消毒领域应用提供一定依据。
【Abstract】 PURPOSE:To explore the changes of physical and chemical properties of hypochlorous acid solution prepared by two production processes under simulated clinical use and storage conditions,and to study the change of the active chlorine concentration of dung enterococcus sterilization effect.METHODS:Hypochlorous acid solution samples for chemical and electrolysis processes were prepared((1)chemical 100 ppm,(2)electrolysis 100 ppm,(3)chemical 200ppm,(4)electrolysis 200 ppm).Under the conditions of simulated clinical use(20-25℃ under light cover storage,open the cover for 5 minutes a day,The pH value,oxidation-reduction potential(ORP) and free available chlorine(FAC) of each solution sample were monitored within 1 month.Suspension quantitative bactericidal test was used to detect the change of the bactericidal value of hypochlorous acid solution against E.faecalis with the decay of FAC.In addition,the above 4 groups of solution samples were kept open or unopened at room temperature and away from light,and FAC was detected at the 6th and 12th month,respectively,to obtain the FAC loss rate of each sample.RESULTS:Under the conditions,the pH value and ORP value of each hypochlorous acid solution were relatively stable,while FAC decreased slowly with the extension of time,and the attenuation degree was less than 20 ppm within 1 month.The FAC loss rates of samples(1)-(4) were 14.05%,19.55%,7.77% and 9.18%,respectively.And the FAC attenuation rate of solution prepared by chemical method was lower than that prepared by electrolytic method(p<0.05).When the FAC of solution at two conventional concentrations(100 ppm and 200 ppm) was reduced by about 20 ppm,the logarithm of bactericidal value against E.faecalis could still be more than 5.00,meeting the bactericidal effect of 99.999% suspension.In addition,the long-term FAC stability of the four groups of sample solutions in the open and unopened state was compared,and the FAC loss rate in the unopened state was only 4.0%-17.6% at 12 months,which was much lower than that in the open state(55.0%-99.0%).CONCLUSIONS:Under simulated conditions,the FAC stability of hypochlorous acid solution with two different production processes is good,and the bactericidal effect can be achieved.The stability of solution FAC prepared by chemical method is better than that by electrolysis method.It provides a basis for the application of hypochlorous acid solution in the field of root canal disinfection.
