百合花粉败育的关键阶段与分子机制研究

【摘要】 为研究百合花粉败育的机理,以亚洲百合花粉败育品种‘小小的吻’与亚洲百合花粉发育正常品种‘小蜜蜂’为材料,通过细胞学观察来检测花粉败育的关键阶段,并在转录水平上测定和分析基因表达信息。结果显示,花粉败育品种‘小小的吻’的小孢子在单核花粉粒阶段发生败育,覆盖在四分体表面的胼胝质未能及时退化。此外,‘小小的吻’的绒毡层细胞较正常品种‘小蜜蜂’相比提前降解,一些涉及绒毡层细胞早期变性和胼胝质生物合成的差异基因和蛋白质表达量发生显著变化。尤其是一些像ARF17、MYB80(103)和AMSL类的转录因子,其表达水平发生显著变化,特别是在败育的单核花粉粒时期发现,负责胼胝质变性的两种β-葡聚糖酶基因(内切-1,3(4)-β-葡聚糖酶,外切-β-葡聚糖酶)显著下调。这些结果表明单核花粉期可能是花粉败育的关键时期。这时期,绒毡层细胞的早期变性导致β-葡聚糖酶基因表达量的下调,β-葡聚糖酶基因表达量的显著下调导致胼胝质降解延迟,小孢子无法和周围进行物质和营养的交换,花粉壁合成受阻,细胞质内含物降解,最终导致花粉败育。

【Abstract】 Our analysis aims to use cytological observation to detect the critical stage of pollen abortion and provides comprehensive gene expression information at the transcriptional level.Pollen abortion of ’ little kiss’ began to take place at the mononuclear pollen stage,and callose covering its microspores failed to degenerate when young pollen released from tetrads.In addition,the tapetum cells of’little kiss’ are degraded in advance compared with the normally developed one.Furthermore,some DEGs and DAPs relate to advanced degeneration of tapetum cells and biosynthesis of callose.Especially some significant changes in the expression levels of some TFs,like ARF17、MYB80(103) and AMSL,were found in abortive stages.In particular,two β-glucanase genes(endo-1,3(4)-beta-glucanase,exo-beta-glucanase) responsible for callose degeneration were significantly down-regulated.Taken together,these results suggested that mononuclear pollen stage may the key period of pollen abortion.Early degeneration of tapetum cells results in significantly down-regulated ofβ-glucanase genes which are responsible for callose degeneration.As a result,the failure in degeneration of callose covering the microspores impedes formation of pollen walls and may leads to pollen abortion.
