PCPA致失眠大鼠松果体组织病理学变化

【摘要】 目的观察对氯苯丙氨酸(parachlorophenylalanine,PCPA)致失眠大鼠松果体病理组织学的影响。方法大鼠随机分为空白对照、PCPA失眠模型,褪黑素(melation,MT)对照组,PCPA模型组及MT对照组大鼠腹腔注射PCPA混悬液450mg/kg,每天一次,连续2d,空白对照组、模型对照组灌胃等体积2%吐温-80碳酸盐溶液,MT阳性对照组灌胃10mg/kg,每天一次,连续7d。ELISA测定血清褪黑素(melatonin,MT)水平,HE染色光镜观察松果体病理结构,透射电镜观察松果体超微结构。结果血清MT水平PCPA模型组显著低于空白对照组(P<0.01),MT对照组显著高于模型组。松果体HE染色光镜病理组织变化:空白对照组松果体细胞形态规则,排列紧密,细胞间界限清晰,胞核完整。PCPA模型组松果体细胞间距变大,细胞界限模糊,排列紊乱,数目明显减少,细胞核向周边移动,空泡变性增多;MT对照组松果体细胞排列紊乱较模型组轻,数目略有减少,空泡变性较少,细胞核固缩减轻,细胞界限较模糊。松果体超微结构变化:空白对照组松果体细胞质中有较多线粒体,溶酶体较少,有较多脂滴;PCPA模型组松果体线粒体多肿胀、嵴断裂、固缩,溶酶体较多,脂滴较少。MT对照组超微结构与空白对照组比较,线粒体肿胀、嵴断裂、固缩较PCPA模型组减轻,溶酶体较少,脂滴较多。结论 PCPA导致大鼠松果体组织和细胞超微结构明显病理损伤,褪黑素改善PCPA致松果体病理损伤。
【Abstract】 Objective The effects on the pathological histopathology of chlorphenylalanine(parachlorophenylalanine,PCPA) in insomnia rats was observed.Methods Rats were randomly divided into blank control and PCPA insomnia model,melatonin(melation,MT) control group,PCPA model group and MT in 450 mg/kg,once a day,2% Twain-80 carbonate solution in 2 d,blank control group,and MT-positive control group in 10 mg/kg,once a day and 7 d.continuously Serum melatonin(melatonin,MT)levels were determined by ELISA,the pathological structure of the pineal body was observed under HE staining,and its ultrastructure was observed under projection electromicroscope.Results The Serum MT level PCPA model group was significantly lower than the blank controls(P <0.01) and the MT control group was significantly higher than the model group.Pathological tissue changes of pineal HE staining photolens:morphological rules of pineal cells,close arrangement,clear inter-cell boundaries and intact nucleus.The PCPA model group of pineal cells became larger,blurred cell boundaries,disordered arrangement,significantly decreased number,nucleus moving to the periphery,and the MT control group was lighter than the model group,slightly decreased number,less bubble degeneration,reduced core consolidation,and blurred cell boundaries.Hyperstructural changes of pineal body:the blank control group had more mitochondrial cytoplasm,less lysosomal cells and more fat drops;PCPA model group mitochondria had more swelling,crest fracture,consolidation,more lysosomal cells,and less fat drops.Compared with MT microstructure and blank controls,mitochondrial swelling,crest fracture and faxation were compared with the PCPA model group,fewer lysosomal and more lipid drops.Conclusion PCPA resulted in obvious pathological damage to the pineal tissue and cell ultrastructure in rats,and melatonin improved the pineal pathology damage caused by PCPA.
