蛇床子催眠活性组分对高架十字迷宫实验焦虑大鼠脑干氨基酸类神经递质的影响

【摘要】 目的观察蛇床子催眠活性组分对焦虑大鼠行为干预效果,并检测脑干氨基酸类神经递质含量,探讨其抗焦虑的可能作用机制。方法建立高架十字迷宫(EPM)实验大鼠焦虑模型,观察蛇床子催眠活性组分对焦虑大鼠行为的影响,酶联免疫吸附测定法(ELISA)测定大鼠脑干内γ-氨基丁酸(GABA)和谷氨酸(GLU)的含量。结果与模型对照组比较,蛇床子催眠活性组分中、高剂量组及地西泮组大鼠进入开臂次数百分比及开臂滞留时间百分比增加(P<0.05);与空白对照组比较,模型组GABA水平显著降低(P<0.01),GLU水平、GLU/GABA值显著升高(P<0.01);与模型对照组比较,蛇床子催眠活性组分中、高剂量组及地西泮组GABA水平显著升高(P<0.05),GLU水平、GLU/GABA值显著降低(P<0.05)。结论蛇床子催眠活性组分有显著的抗焦虑作用,其作用机制与增加脑干GABA抑制性与降低GLU兴奋性氨基酸类神经递质,调节二者比例平衡有关。
【Abstract】 Objective To investigate the effects of Cnidii Fructus active hypnotic components(CHC) on behavioral intervention in rats with anxiety,and to examine the levels of amino acid neurotransmitters in the brain stem to further determine its potential mechanism of anti-anxiety.Methods The EPM anxiety ratmodels were established.The effects of(CHC) on behavioral intervention in rats with anxiety were determined.The levels of gamma-aminobutyric acid(GABA) and glutamate(GLU) in the brain stem were examined by enzyme-linked immunosorbentassay(ELISA).Results Compared with the model group,the percentages of open-arms entries and open-arms length in the medium and high-dose CHC groups and diazepam group increased(P<0.05).Compared with the blank control group,the GABA level in the model group significantly decreased(P<0.01),whereas the GLU level and the ratio of GLU/GABA significantly increased(P<0.01).Compared with the model group,the GABA level in the medium and high-dose CHC groups and diazepam group significantly increased(P<0.05),whereas the GLU level and the ratio of GLU/GABA significantly decreased(P<0.05).Conclusion CHC shows significant anti-anxiety effect,and its mechanism may be related to the increase of GABA inhibition in brain stem,decrease of GLU excitatory amino acid neurotransmitter,and regulation of the balance of GLU/GABA ratio.
