甲壳素纳米纤维/玉米醇溶蛋白纳米粒子稳定的O/W型Pickering乳液调控三丁酸甘油酯脂水解过程

【摘要】 短链脂肪酸尤其是丁酸对人体肠道的有益作用已经得到广泛的认可。为了使人体结肠和外周组织获得足够水平的丁酸,服用丁酸前体-三丁酸甘油酯是一种替代给药途径。然而三丁酸甘油酯本身较差的溶解度和极易被消化的特性极大地限制了其在功能食品中的应用。本文构建甲壳素纳米纤维与玉米醇溶蛋白稳定的O/W型Pickering乳液用于递送三丁酸甘油酯,探究纳米纤维/蛋白纳米粒子比例对乳液稳定性、包封三丁酸甘油酯能力以及负载的三丁酸甘油酯消化特性的影响。结果表明,当甲壳素纳米纤维/蛋白纳米粒子含量比为1:5时,三丁酸甘油酯为油相的乳液油脂完全消化所需时间为70 min,远高于乳清分离蛋白作为乳化剂稳定的乳液完全消化所需的时间(20 min)。进一步采用肉桂醛界面交联法修饰Pickering乳液,消化结果表明修饰后三丁酸甘油酯完全消化的时间延长至120min,有效延缓三丁酸甘油酯的脂水解过程。油脂消化控制型乳液的开发有望用于保护递送易水解功能性油脂通过胃肠道,靶向进入结肠部位。
【Abstract】 The beneficial effects of short-chain fatty acids,especially butyrate,on human gut have been widely recognized.Tributyrin,a precursor of butyrate,is always used as an alternative route for oral administration in order to deliver butyrate to the human colon and peripheral tissue.However,the poor solubility and highly digestibility of tributyrin greatly limit its application in functional food.In this work,O/W Pickering emulsions were stabilized by chitin nanofibers/zein nanoparticles complex.The influence of chitin nanofibers/zein nanoparticles ratios on the stability of emulsion,the encapsulation efficiency and digestibility of tributyrin was investigated.The results showed when the mass ratio of chitin nanofibers/zein nanoparticles was1:5,it took 70 min to reach the complete digestion of tribyturin in emulsions.In comparison,the time required for complete digestion of tributyrin in whey protein isolate-stabilized emulsion was 20 min.Furthermore,cinnamaldehyde was used to modify the interfacial layer of Pickering emulsions.In vitro digestion results indicated that the time to complete digestion of tributyrin was extended to 120 min after modification,effectively delaying its lipolysis process.The development of lipid digestion-controlled emulsions is promising to protect the delivery of highly-digestible functional lipids through the gastrointestinal tract,targeting them to the colonic site.
