外周全血环状RNA5430是肺腺癌新的生物标志物

【摘要】 目的研究肺腺癌患者外周全血中环状RNA5430 (circRNA5430)的表达情况,为肺腺癌的早期筛查提供新的生物标志物。方法通过高通量测序,筛选出肺腺癌外周血circRNA差异表达谱;在20例肺腺癌患者和10例正常人群中,采用实时荧光定量逆转录聚合酶链反应(qRT-PCR),验证差异表达的circRNA5430。采用Spearman相关分析,对circRNA5430与临床变量的相关性进行评价。绘制受试者操作特征曲线(ROC),评价circRNA5430对肺腺癌的诊断效能。结果与对照组相比,肺腺癌患者的外周血circRNA5430表达明显上调,与疾病活动(CEA、CA199、CA50)显著正相关;ROC曲线显示,ROC曲线下面积AUCROC为:0.75 (95%置信区间:0.567-0.928,P<0.05),敏感度为0.65,特异性为0.90,表明外周血中的circRNA5430可以区分肺癌患者和对照组。结论肺腺癌外周血中的circRNA5430可能是肺腺癌患者早期诊断的潜在的生物标志物。

【Abstract】 Objective:This study aimed to determine the expression of circular RNA5430(circRNA5430) in peripheral whole blood(PWB) from lung adenocarcinoma(LA) patients to identify novel biomarkers for LA screening.Methods:We initially performed circRNA expression profile in PWB by high-throughput sequencing.We then confirmed circRNA5430 changes in PWB from 20 LA patients and 10 healthy controls using real-time quantitative reverse transcription-polymerase chain reaction.Spearman’ correlation test was performed to assess the correlation of circRNA5430 and clinical variables.The receiver operating characteristic(ROC) curve was drawn to evaluate diagnostic efficiency for LA.Results:We confirmed that PWB circRNA5430 was significantly upregulated in LA patients when compared with healthy controls.Among these LA patients,we detected significant correlation between PWB circRNA5430 and disease activity.ROC analysis showed that PWB circRNA5430 had an area under the curve of 0.75(95% CI 0.567-0.928,P <0.05) to discriminate LA patients from controls with sensitivity of 0.65 and specificity of 0.90.Conclusions:This study revealed that upregulated PWB circRNA5430 in LA may serve as a potential biomarker for LA patient diagnosis.
