老年人大便性状与肠道菌群的相关分析

【摘要】 目的:比较老年人大便性状的不同与肠道菌群间的联系。方法:选取便秘组、便溏组、对照组共32例,留取新鲜粪便样本及血样,采用16SrRNA测序技术对V4区进行粪便微生物DNA测序,分析肠道菌群物种的丰度和分布;用ELISA法检测炎症因子TNF-α、IL-10、IL-17、IL-22、LTB-4、PGE2,分析各组间炎症因子的差异。结果:在三组间,菌群的多样性(simpson和shannon)差异具有统计学意义,丰富度(Ace和Chao1)差异性无统计学意义;在属的水平,Collinsella(x2=6.599,P <0.05)、Anaerostipes(x2=8.707,P <0.05)、Bilophila(x2=7.354,P <0.05)、Anaerotruncus(x2=6.179,P <0.05)及Megasphaera(x2=6.095,P <0.05)在三组间的丰度差异具有统计学意义,并且Collinsella及Bilophila丰度与大便稠度呈显著相关(r=0.449,P <0.01;r=0.455,P <0.01);但各炎症因子在三组间的差异无统计学意义。结论:在老年人中,大便性状的不同与肠道菌群的变化有关,通过调节肠道菌群来改善老年人胃肠功能紊乱值得进一步研究。更多还原

【Abstract】 Objective: To investigate the link between gut microbiota and stool consistency of the older.Methods: The 16S rDNA high-throughout sequencing of fecal sample was carried out to analysis the difference of abundance of gut flora in three groups which include constipation group, normal group, and loose stool group.The ELISA technique was adopted to detect the inflammatory cytokines, such as TNF-α、IL-10、IL-17、IL-22、LTB-4、PGE2. Results:There is a significant difference of the bacterial diversity(Simpson index and Shannon index) in three groups, but not for richness(Ace index and Chao1 index). At the level of genus, the obvious difference is observed in three groups on Collinsella(x2=6.599,P < 0.05),Anaerostipes(x2=8.707,P < 0.05),Bilophila(x2=7.354,P < 0.05),Anaerotruncus(x2=6.179,P < 0.05)and Megasphaera(x2=6.095,P < 0.05). Besides, the abundance of Collinsella and Bilophila are strongly correlated with stool consistency(r=0.449,P < 0.01;r=0.455,P < 0.01). But no significant difference is observed about inflammatory cytokines. Conclusion: The old people’s stool consistency is associated with the variation of gut microbiota. It is deserved further study to improve the gastrointestinal dysfunction in the elderly through the regulation of intestinal microflora.更多还原
