S/O/W钙-脂质微球构建与稳定性研究

【摘要】 碳酸钙(CaCO3)因其钙含量高、价格低廉常用做强化钙,但其悬浮稳定性差,易产生沉淀。本研究基于S/0/W型乳液技术构建碳酸钙-脂质微球,以物理稳定性和包埋效果为指标,探究构建碳酸钙-脂质微球的最佳条件,解决难溶性钙盐在液态食品中的分散稳定性。首先对比了CaCO3在以大豆油或猪油为O相制备的S/O初乳液中的分散效果,结果表明:猪油在低温时,因其固体脂肪指数较大能够表现出较高的黏度,从而使碳酸钙在均匀分布到O相后不易沉淀。在此基础上,采用高速剪切制备终乳液,比较W相海藻酸丙二醇酯(PGA)与黄原胶(XG)复配时不同质量比、不同总物质浓度、pH值所构建的碳酸钙-脂质微球物理稳定性及包埋效果,结果表明:在PGA与XG质量比为3:7,总物质浓度为0.8%,pH=6时,微球平均粒径为(8.02±0.27)μm,Zeta-电位为(-45.92±0.96) mV,微流变出现平台区域,具有良好的物理稳定性,同时微观结构(CLSM)表明CaCO3被较好的包埋在体系中。为研制矿物质新型传递系统提供了理论和技术基础。
【Abstract】 Calcium carbonate(CaCO3) is commonly used as reinforcing calcium due to its high calcium content and low price,but its suspension stability is poor,which is prone to precipitate.In this study,calcium carbonate-lipid microspheres were constructed based on S/O/W emulsion technology.The physical stability and embedding effect were used as indicators to explore the optimal conditions for the construction of calcium carbonate-lipid microspheres,so as to solve the dispersion stability of insoluble calcium salts in liquid food.Firstly,the dispersion effect of CaCO3 in the S/O primary emulsion prepared with soybean oil or lard as O phase was compared.The results showed that lard at low temperature could exhibit high viscosity due to its large solid fat index,so that calcium carbonate was not easy to precipitate after evenly distributed to the O phase.On this basis,the final emulsion was prepared by high-speed shearing.The physical stability and embedding effect of calcium carbonate-lipid microspheres were compared when the W phase was composed of propylene glycol alginate(PGA) and xanthan gum(XG) with different mass ratios,different total material concentrations and pH values.The results showed that when the mass ratio of PGA and XG was 3:7,the total material concentration was 0.8%,and pH at 6,the average particle size of the microspheres was(8.02±0.27) μm,and the Zeta-potential was(-45.92±0.96) mV.The micro-rheology appeared a platform area,which had good physical stability.At the same time,the microstructure(CLSM) indicated that CaCO3 was well embedded in the system.It provides a theoretical and technical basis for the development of new mineral transfer system.
