富硒大豆肽SSeCAHK对过氧化氢诱导的HepG2细胞氧化应激的保护作用

【摘要】 本研究旨在从富硒大豆肽中制备含硒大豆抗氧化肽,并研究其在体外氧化应激中的作用。富硒大豆肽通过凝胶过滤被分离成5个组分(F1~F5),并通过DPPH、ABTS和OH-自由基清除实验确定抗氧化活性最高的组分。进一步通过阴离子交换层析分离抗氧化活性最高的组分F4,得到F4-1~F4-5亚组分,通过DPPH、ABTS和OH-自由基清除试验评价其抗氧化活性。在F4-1亚组分中鉴定出序列为Ser-SeC-Ala-His-Lys(SSeCAHK)的含硒大豆抗氧化肽,并化学合成。测定SSeCAHK对过氧化氢诱导的HepG2细胞氧化应激的预防作用。SSeCAHK(0.13~0.50 mg/mL)预处理HepG2细胞2 h后,细胞内活性氧清除能力增强,同时防止了还原性谷胱甘肽含量下降和丙二醛含量增加。因此,SSeCAHK处理可改善过氧化氢诱导的HepG2细胞的氧化应激反应,表明SSeCAHK可作为膳食补充天然抗氧化功能材料,具有非常大的潜力。
【Abstract】 This study aimed to prepare selenium(Se)-containing soybean antioxidant peptides from Se-enriched soybean peptides(SSP) and determine their roles in oxidative stress in vitro.In this work,the SSP was separated into five fractions(F1 to F5).Fraction F4,displaying the highest antioxidative activity,was further separated,and sub-fractions F4-1 to F4-5 were selected for antioxidative activity evaluation using DPPH,ABTS,and OH-radical scavenging assays.The Se-containing antioxidative peptides with sequence Ser-SeC-Ala-His-Lys(SSeCAHK) were identified in sub-fraction F4-1 and chemically synthesized.This Se-containing pentapeptide showed a preventive effect against hydrogen peroxide(H2 O2)-induced oxidative stress in HepG2 cells.Pretreating the cells for 2 h with SSeCAHK(0.13-0.50 mg/mL) induced strong intracellular,reactive oxygen species(ROS) scavenging activity while preventing a decrease in reduced glutathione(GSH) and an increase in malondialdehyde(MDA).Therefore,SSeCAHK treatment improved H2 O2-induced oxidative stress in HepG2 cells,demonstrating the significant potential of SSeCAHK as a natural antioxidative functional material for dietary supplementation.
