砀山纯梨膏指纹图谱的建立及其相似度评价

【摘要】 目的:运用高效液相色谱(HPLC)法建立不同产梨区砀山纯梨膏的指纹图谱,为其质量控制和鉴定方法提供参考依据。方法:以选自砀山县6个经典产梨区酥梨自制的纯梨膏为实验材料,采用纯甲醇回流提取2次,并用岛津C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇和0.1%甲酸水进行梯度洗脱,柱温30℃,流速1.0 mL/min,检测波长280 nm,并对其测定结果采用指纹图谱相似度软件进行数据分析,通过主成分分析判别市售非梨膏及非纯梨膏产品。结果:6个产区纯梨膏指纹图谱相似度在0.987~0.996,且6个梨膏产品的酚酸、黄酮和三萜类成分含量均无显著性差异(P<0.01),单量范围分别为3.144~3.489 mg/g、0.137~0.229 mg/g和1.939~2.272 mg/g;主成分分析结果表明,6个纯梨膏产品具有较高的相似性,与市购的非梨膏产品及非纯梨膏产品有明显的区别。结论:砀山县6个经典产梨区纯梨膏在品质上均无显著差异,所建立的HPLC指纹图谱可以准确判别市售非梨膏及非纯梨膏产品。

【Abstract】 Objectives:The fingerprint of pure pear paste made from Dangshan pear in different pear producing areas were established by high performance liquid chromatography(HPLC) to provide a reference for the quality control and scientific application of pure pear paste.Methods:Pure pear paste from six classical pear-producing areas in Dangshan County was selected as the experimental material.Pure methanol was used for twice reflux extraction,and Shimadzu C18(4.6 mm ×250 mm,5 μm) column was used for gradient elution with methanol and 0.1% formic acid water.Column temperature was 30℃,flow rate was 1.0 mL/min,and detection wavelength was 280 nm,fingerprint similarity software was used for data analysis,and principal component analysis was used to distinguish non-pear paste products and non-pure pear paste products.Result:The similarity of pure pear paste from 6 producing areas ranged from 0.987 to 0.996,and there was no significant difference in the contents of phenolic acids,flavonoids and triterpenoids among the six pear paste products(P <0.996).The unit quantities ranged from 3.144 to 3.489 mg/g,0.137 to 0.229 mg/g and 1.939 to 2.272 mg/g,respectively.The results of principal component analysis show that the six pure pear paste products have high similarity,which is obviously different from the non-pear paste products and non-pure pear paste products purchased in the market.Conclusion:There is no significant difference in the quality of pure pear paste from six classical pear producing areas in Dangshan County.The HPLC fingerprint can accurately distinguish non-pear paste and non-pure pear paste products.
