大刺鳅人工繁殖与胚胎发育研究

【摘要】 为探究大刺鳅人工繁殖和胚胎发育特性,选择人工培育的三冬龄大刺鳅做亲本,在28℃~29℃水温下,分两次注射绒毛膜促性腺激素、促黄体素释放激素A2和地欧酮混合物,两次注射间隔时间12h,第二次注射后12h开始人工授精。大刺鳅受精卵为沉性卵,具粘性,受精卵吸水膨胀后,卵径为1.94±0.05mm。结果显示,大刺鳅催产率为99.20%,受精率为82.77%,孵化率为77.20%,雌鱼平均怀卵量约3036粒/尾;受精卵在水温28℃~29℃、溶解氧5mg/L以上、p H值7.2~7.8的条件下,胚胎发育历经受精卵、胚胎隆起、卵裂期(2细胞、4细胞、8细胞、16细胞、多细胞期、桑葚期)、囊胚期(囊胚早期、囊胚中期、囊胚晚期)、原肠胚期(原肠早期、原肠中期、原肠晚期)、神经胚期(神经胚期、胚孔封闭期)、器官形成期(肌节出现期、视泡期、尾芽期、肌肉效应期、心跳期)、出膜期等8个阶段22个时期,总历时约50.5h,总积温约1454.4℃·h。初孵仔鱼平均全长6.52±0.05mm。

【Abstract】 In order to break through the large-scale rapid and efficient breeding technology of Mastacembelus armatus,And the morphological characteristic of embryonic development was observed in detail under the water temperature of 28℃~29℃.The fertilized eggs of Mastacembelue armatus were acquired by artificial insemination through two injections of HCG,LRh-A2 and DOM mixture.The results showed that the rate of oxytocin was 99.20%,fertilization rate was 82.77%,and hatching rate was 77.20%.The average number of eggs in the female parent is about 3036 per tail.Under the water temperature of 28~29℃,it took about 50.5h during the embryonic development of Mastacembelue armatus and the total accumulative temperature was about 1454.4℃·h.The embryonic development process could be divided into fertilized egg stage,the uplift of blastoderm stage,cleavage stage(2 cells stage,4 cells stage,8 cells stage,16 cells stage,multicellular stage,and mulberry stage),blastula stage(early,middle and late stage),gastrula stage(early,middle and late stage),nerve embryonic stage(nerve embryonic stage,embryo hole closed stage),organ formation stage(sarcomere appearance stage,visual bubble stage,caudal bud stage,muscle response stage,heartbeat stage),hatching stage,and 22 developing phases.
