【摘要】 目的 :评价氧化锆表面聚乙烯亚胺(PEI)涂层接枝绿原酸(CGA)-壳聚糖(CS)复合物对骨结合促进的效果。方法 :以壳聚糖包裹绿原酸,再用PEI将其接枝于氧化锆表面,建立生物活性涂层。采用扫描电子显微镜(SEM)、X射线光电子能谱(XPS)、接触角计对涂层进行表征分析。利用具有高成骨分化潜能的小鼠前成骨细胞系MC3T3-E1评价涂层的促粘附及成骨诱导作用。结果 :XPS证实PEI及绿原酸复合物接枝于氧化锆植体表面;接触角显示该涂层增强了氧化锆表面的亲水性;激光共聚焦显微镜发现该涂层能促进细胞的增殖及粘附;蛋白质免疫印迹法显示OCN、Runx2等成骨指标上调,提示该涂层的早期促成骨作用。结论 :氧化锆植体表面聚乙烯亚胺涂层接枝绿原酸-壳聚糖复合物具有促进氧化锆表面成骨的作用。更多还原

【Abstract】 Objective: To evaluate the effect of polyethylenimine(PEI) coating on the surface of zirconia grafted with chlorogenic acid(CGA)-chitosan(CS) complex on the promotion of osseointegration. Methods: CS wrap the the CGA and graft it onto the surface of zirconia implants with PEI to build a biologically active coating. Using scanning electron microscope(SEM), X-ray photoelectron spectroscopy(XPS), water contact angle measurement and other experiments to analyze the coating’s characterization. Pre-osteoblastic MC3T3-E1 cells with high osteogenic differentiation potential was used to evaluate the adhesion promotion and osteoinduction effects of the coating. Results: XPS confirmed that PEI and chlorogenic acid complexes was grafted on the surface of zirconia implants; Water contact angle shows that good hydrophilic coatings are formed on the surface of zirconia; laser confocal microscopy confirms that chlorogenic acid can promote cell proliferation and adhesion; Western blotting showed that OCN, Runx2 and other osteogenic indicators were up-regulated, confirming that chlorogenic acid can promote early osteogenesis. Conclusions: This study confirmed that chlorogenic acid can increase the biological activity of zirconia surface to promote bone integration, which provides a new idea for the potential application of zirconia surface activity treatment.更多还原
