长毛兔KRT17基因启动子甲基化对产毛性能的影响

DNA methylation of KRT17 gene promoter and its influence on wool production traits in wool rabbits

【摘要】 为探究家兔KRT17基因启动子DNA甲基化水平与产毛性能的相关性,通过软件预测KRT17基因启动子CpG岛,采用亚硫酸氢盐测序(BSP)法分析KRT17基因启动子CpG岛的甲基化水平。结果发现,兔KRT17基因启动子区存在3个CpG岛(Cp GⅠ-Ⅲ),其中CpGⅠ位于-1567~-1403bp,包含16个CpG位点,在高/低产毛组中的整体甲基化水平分别为43.92%和61.37%,差异显著(P<0.05),且-1539bp位点的甲基化水平与产毛量显著相关(P<0.05)。CpGⅡ位于-1186~-1053bp,包含10个CpG位点,在高/低产毛组中的整体甲基化水平分别为10.00%和11.21%,无显著性差异(P>0.05),且没有与产毛量显著相关的CpG位点。CpGⅢ位于-97~+120bp,包含18个CpG位点,在高/低产毛组中的整体甲基化水平分别为43.92%和61.37%,差异不显著(P>0.05),但-53、-51、-38、-33bp位点的甲基化水平与产毛量显著相关(P<0.05)。进一步结合转录因子结合位点预测分析发现,CpGⅢ中的-38、-33bp位点处于转录因子SP1的结合位点,提示甲基化修饰对产毛量的影响可能是由影响SP1的结合导致。

【Abstract】 To investigate the correlation between DNA methylation levels and wool production performance of rabbit KRT17 gene promoter,the CpG island was predicted by software,and the degree of methylation was detected by bisulfite sequencing PCR(BSP).The results showed that there were three CpG islands(Cp GⅠ-Ⅲ)in the promoter region of KRT17 gene,in which CpGI was located in-1567~-1403 bp,containing 16 CpG sites,the overall methylation levels in the high/low hair group were 43.92% and 61.37%(P<0.05),and the methylation locus of-1539 bp was significantly correlated with wool production(P<0.05).CpGII was located in-1186~-1053 bp and contained 10 CpG sites,the overall methylation levels in the high/low hair group were 10.00% and 11.21%(P>0.05),and there is no CpG site that is significantly associated with wool production.CpGIII was located in-97~+120 bp,containing 18 CpG sites,The overall methylation levels in the high/low hair production group were 43.92% and 61.37%(P>0.05),the methylation loci of-53,-51,-38,and-33 bp were significantly correlated with wool production(P<0.05).Further analysis combined with transcription factor binding site prediction revealed that the-38,-33 bp locus in CpGIII is the binding site of transcription factor SP1,suggesting that the effect of methylation modification on wool production may be caused by the combination of SP1.
