基于鲁米诺电致化学发光的生物传感器研究

【摘要】 鲁米诺电化学发光体系是基于电化学氧化成鲁米诺生成自由基,所生成的自由基不稳定,再进一步被一些氧化剂氧化生成化学发光。ECL分析方法具有灵敏度高,线性范围宽和仪器设备简单等优点[1]。传统的鲁米诺ECL应用多在强碱性介质中进行,但是在中性,弱碱性等介质利于生物分子保持活性的环境中,鲁米诺的ECL发光极弱[2]。为了保持生物分子活性,考虑利用合理措施实现鲁米诺在中性,弱碱性介质环境中的ECL曾敏,对其发展和应用都有很高的学术和应用价值。由于过度金属氧化物Co3O4纳米线经过NaBH4还原后,其比表面积增大,导电性能增强,催化性能增强,制造成本低等优点[3],与鲁米诺通过金属配位键形成Co3O4纳米线/鲁米诺复合物,且还原型Co3O4纳米线表面氧空位能够活化氧气形成超氧阴离子与鲁米诺自由基反应,有效增强鲁米诺的ECL.在最佳实验条件下,我们制备了鲁米诺ECL免疫传感器,用于检测癌胚抗原(CEA),线性范围是0.005-500 ng/ml,且检出限为0.005 ng/mL(S/N=3)[4]

【Abstract】 The luminol electrochemiluminescence system is based on electrochemical oxidation to luminol to generate free radicals.The generated free radicals are unstable and are further oxidized by some oxidants to generate chemiluminescence.The ECL analysis method has the advantages of high sensitivity,wide linear range,and simple equipment.The traditional ECL application of luminol is mostly carried out in a strongly alkaline medium,but in an environment where neutral,weak alkaline,and other media are conducive to maintaining the activity of biomolecules,luminol’s ECL luminescence is extremely weak.To maintain the activity of biomolecules,consider the use of reasonable measures to realize the ECL intensity increase of luminol in a neutral and weakly alkaline medium environment,which has high academic and application value for its development and application.Due to the transition metal oxide Co3O4 nanowires were reduced by NaBH4,its specific surface area is increased,the conductivity is enhanced,the excellent catalytic performance is enhanced,and the manufacturing cost is low.It forms a Co3O4 nanowire/luminol composite through the metal coordination bond with luminol,and the oxygen vacancies on the surface of reduced Co3O4 nanowires can activate oxygen to form superoxide anions to react with luminol free radicals,effectively enhancing the ECL of luminol.Under the best experimental conditions,we have prepared a luminol ECL immunosensor for cancer detection Embryonic antigen(CEA),the linear range is 0.005-500 ng/ml,and the detection limit is 0.005 ng/mL(S/N=3).
