基于荧光-磁性-催化纳米球信号放大的H9N2禽流感病毒双模态检测

【摘要】 H9N2禽流感病毒(H9N2 AIV)的爆发每年带来高的死亡率和巨大的经济损失,因此建立灵敏和可靠的检测方法对于实现及时诊断和治疗至关重要[1],[2]。本文借助荧光-磁性-催化纳米球(FMCNs)作为信号标签和碱性磷酸酶(ALP)诱导金属化作为信号放大策略构建了一种双模态的免疫分析平台用于H9N2 AIV灵敏检测(Fig.1)。凭借抗原抗体特异性反应,目标物H9N2 AIV能够被FMCNs捕获形成H9N2 AIV/FMCNs复合物,在外加磁场下,H9N2 AIV/FMCNs复合物能够从复杂样品中分离出来并被多克隆抗体(p Ab)标记的三组分修饰电极捕获形成三明治夹心结构。FMCNs好的荧光性能能够提供荧光信号输出,而ALP诱导金属化则可以提供好的电化学信号的输出,因此能够实现H9N2 AIV的双模态检测。

【Abstract】 The outbreak of H9N2 avian influenza virus(H9N2 AIV) brings high mortality and huge economic losses every year.Sensitive and reliable detection methods are essential to timely diagnosis and treatment.Herein,a dual modality immunoassay is proposed for H9N2 AIV detection by employing fluorescent-magnetic-catalytic nanospheres(FMCNs) as labels and alkaline phosphatase(ALP)-induced metallization as a signal amplification strategy.The targets could be captured by FMCNs from complex samples to form target/FMCN complexes in virtue of the specific reaction of antigen-antibody.Under an external magnetic field,target/FMCN complexes were separated from complex samples and captured by the polyclonal antibody(pAb) labeled triple-component modified ITO electrode to form sandwich structures.The excellent fluorescence properties of FMCNs enable fluorescence modality readout and amplified electrochemical modality readout can be obtained through ALP-catalyzed silver deposition,thus achieving the dual-modality detection of H9N2.
