基于卵黄抗体标记MOF探针的电化学免疫传感器检测致病菌研究

【摘要】 对食源性致病菌污染检测迫切需要开发快速简便的现场分析方法。我们以金黄色葡萄球菌(S. aureus)为致病菌[1]代表,用灭活S. aureus免疫蛋鸡并提取卵黄抗体(Ig Y),并证明其可以高效且特异性地捕获S.aureus。将Ig Y和UiO-66构建复合MOF探针和电化学免疫传感器。其对S. aureus检测范围为10~109 CFU/mL,检测下限为3 cfu/mL,检测了养殖虾体内的S. aureus,获得的结果和标准ELISA一致,但是检测时间大大缩短,检测灵敏度提高100倍。这说明该法非常适合于食品中金黄色葡萄球菌的快速分析
【Abstract】 The food-borne pathogenic staphylococcus aureus(S. aureus) pollution is one important cofactor affecting the quality and safety of foods. It is urgent to develop some rapid, simple and on-site analytical methods to detect them. In the study, the egg yolk antibody(IgY) was extracted and modified on UiO-66 to fabricate MOF probe and the electrochemical immunosensors for S. aureus measurement. The sensor was employed to detect S. aureus in farmed shrimp. The results were consistent with standard ELISA result, which demonstrated that the detection time is greatly shortened and the detection sensitivity is increased by 1,000 times. All results showed that the assay is suitable for rapid analysis of S. aureus in food
