Cascade Decarboxylation and Cyclization towards 3-Difluoromethylene-containing Chroman-4-one Derivatives

【摘要】 The chroman-4-one scaffold is important structure motif in many natural products,medicinal compounds and biological compounds.1 Especially,introducing functional groups at the 3-position of the chroman-4-one scaffold can greatly improve their activity.Hence,the development of approaches to synthesis these 3-position functionalized chroman-4-ones has attracted much attention.2 However,only a few examples have been reported for the synthesis of 3-fluorine-containing chroman-4-one derivatives.3 Therefore,it would be highly desirable to introduce the ArCF2 moiety into the chroman-4-one derivatives to promote their application in drug discovery.Herein,we describe an effective and transition-metal-free cascade decarboxylation and cyclization of α,α-difluoroarylacetic acids and o-(allyloxy)arylaldehydes for biologically important3-difluoromethylene-containing chroman-4-one derivatives under mild conditions(Scheme 1).
