基于DNA组装体的金纳米阵列构筑

【摘要】 脱氧核糖核酸(DNA)分子是结构复杂的有机化合物,具有可预测和可编程的分子间相互作用,由此可构筑各种各样的纳米组装体。2006年,Rothemund提出了一种新的DNA自组装方法——DNA折纸术,将长的单链DNA与一系列设计好的DNA短片段进行碱基互补配对,利用"一锅煮"的方法快速、精准地构建高度复杂的纳米结构。本文设计并构筑了一系列基于DNA组装体的金纳米阵列,将两个不同的环状DNA模板(均含有63个不同的特定碱基序列)经滚环扩增反应生成两条含有多个重复序列的长链DNA(分子量约10000 bp),进而与42个碱基短链进行碱基互补配对,得到了相较于单条DNA长链更具有刚性的DNA纳米阵列。借助42个碱基短链的末端巯基,与金纳米颗粒进行结合,实现了纳米金阵列的有序组装。上述结构有望应用于生物传感器、纳米粒子自组装、载药等方面。
【Abstract】 Deoxyribonucleic acid(DNA) molecules can be used to construct various nanostructures for their predictable and programmable intermolecular interaction. In 2006, Rothemund proposed a new method of DNA self-assembly, DNA origami. Long single strands of DNA are paired with designed short pieces of DNA due to their complementary bases. Through a "one-pot" method, complex nanostructures can be quickly and accurately constructed from the above mixture. The purpose of this paper was to design and construct a series of nano-arrays based on DNA self-assembly. By roll circle amplification(RCA) reaction, two different circular DNA templates(both including 63 different specific bases sequence) were used to generate two long DNA chains containing more repetitive sequences( about 10000 bp). Then, the two long DNA chains were mixed with short DNA chains containing 42 bases to form DNA nano-arrays, which have much higher rigidity than one single DNA chain. Finally, gold nanoparticles can be combined with the DNA nano-arrays for their interaction with the mercapto groups on the 5’ end of the short DNA chain containing 42 bases. Thus, the gold nano-arrays were obtained, which have potential applications in biosensors, self-assembly of nanoparticles, drug delivery.
