酒石酸盐对Bola型短肽自组装行为的调控

【摘要】 由于良好的生物相容性和丰富的自组装形貌,多肽自组装已发展成为生物化学交叉学科的新增长点,是制备新材料和产生新功能的重要手段。为实现组装体形貌和尺寸的调控,研究人员常通过改变分子结构(氨基酸的手性、排列次序、端基等)和外部因素(溶剂、pH值、温度等)来调控不同非共价键力,进而调控组装体形貌。本论文报道了一种利用酒石酸盐来调控纳米管管壁厚度的简便有效方法。前期针对Bola型短肽Ac-KI3VK-NH2的研究表明,Ac-KI3VK-NH2可以自组装形成直径约40 nm的纳米管,管壁厚度为单分子层。当将该多肽与含有两个羟基的酒石酸钠以不同比例混合时,发现纳米管的壁厚随着酒石酸钠(STA)比例的增加从单层转变为双层。在此基础上,我们尝试考察了双层管形成的动态过程,为解释双层纳米管的形成过程和形成机制提供了思路。为进一步阐明酒石酸盐结构对双层管形成的影响,我们分别用含有一个羟基的苹果酸钠(SMA)以及不含羟基的丁二酸钠(SSA)作为对比分子,发现羟基的数量对纳米管的壁厚起决定性作用。接着我们考察了酒石酸盐的手性和带电状态对Ac-KI3VK-NH2形成管的壁厚的影响,发现酒石酸盐的手性对双层管的管壁厚度无明显影响,而其带电状态能够显著影响管壁的厚度。上述研究工作的开展为多肽组装体形貌的调控提供了重要思路。

【Abstract】 As for the superior properties, such as good biocompatibility and abundant self-assembly morphologies, peptides self-assembly has become a new growth point in the biochemistry interdisciplinary, which is always regarded as important tool for the preparation of new materials with new functions. In order to regulate the aggregate morphology, researchers can change both the molecular structure of the peptide(chiral, sequence, terminal group, etc.) and the external conditions(solvent, pH, temperature, etc.).This paper reports a new method for regulating the thickness of nanotube walls by the introduction of tartrate into the system. Our previous studies have shown that the amphiphilic short peptide Ac-KI3 VK-NH2 with Bola-Like structure can self-assemble to form nanotubes with a diameter of about 40 nm(the monolayer of the tube wall), When the peptide was mixed with sodium tartrate containing two hydroxyl groups in different ratios, it was found that the wall thickness of the nanotubes changed from single layer to double layer with the increase sodium tartrate(STA) content. Then, we try to investigate the dynamic process for the double layer nanotubes formation, which can help us better understand the formation process and mechanism of double layer nanotubes. To further clarify the influence of tartrate structure on the formation of double-layer tubes, we used sodium citrate(SMA) containing a hydroxyl group and sodium succinate(SSA) without hydroxyl as a comparative salt molecule, and found that the amount of hydroxyl groups plays a decisive role in the wall thickness of the nanotubes, which explains the role of hydroxyl groups in the assembly process of peptides and provides a new research idea for us to directionalally regulate the wall thickness of nanotubes
