单十二烷基磷酸钠/戊醇/水三元体系囊泡的构筑及其在原细胞膜模拟中的应用

【摘要】 目前,大多数研究者将脂肪酸体系作为原始细胞膜模型,然而,非脂肪酸类单一单链表面活性剂囊泡,特别是将其作为一种新的原始细胞膜模型了解甚少。因此,选取具有原始相关性的单链表面活性剂十二烷基磷酸钠(SDP)作为原细胞膜组分,从而构筑新型的原始细胞膜模型。在正戊醇(NPT)的存在下,SDP在水中的单相区显著增大,相关的相行为变化通过TEM、DLS、SAXS、AFM、CLSM、电导率法等进行了表征。值得一提的是,SDP/NPT囊泡在其p Ka附近能够自发形成,且形成的囊泡双层膜具有极端条件下的稳定性(时间稳定性、热稳定性、冻融稳定性、抗稀释性)以及尺寸选择渗透性。借助于SDP/NPT囊泡的上述原细胞膜特性,将其作为酶级联反应器,模拟了原细胞群落间化学信号的传递过程。
【Abstract】 At present, fatty acid are considered as the protocell membranes model system in most system in most cases. But for the case of non-fatty acid single-chain amphiphiles(SCA) vesicle, we know very little about them, especially the study of SCA vesicle as a novel model of protocell membranes. Therefore, we select sodium monododecyl phosphate(SDP), a primordial relevant single-chain amphiphiles, as constituent of protocell compartments model. The addition of n-pentanol(NPT) into the SDP/H2O mixed system markedly enlarged the field of the isotropic phase. The relevant phase behavior has been extensively characterized by transmission electron microscopy(TEM), dynamic light scattering(DLS), small angle X-ray scattering(SAXS), atomic force microscopy(AFM), laser scanning confocal microscopy(LSCM), and conductivity. It is worth pointing out that SDP/NPT vesicles formed spontaneously nearby its p Ka, and the vesicular bilayer membrane has the stability under extreme environment(long-term storage, high temperature, freeze-thawing, dilution resistance) and selective permeability. Based on the primitive cell membrane characteristics of SDP/NPT vesicles, used it as an enzyme cascade reactor, mimicking the prebiotic chemical signaling communication scenario on the early Earth.
