具有良好生物相容性和优异细胞保护功能的磁性富勒醇/铁微胶囊

Magnetic and Biocompatible Fullerenol/Iron Microcapsules with Favorable Cell Protection in Oxidative Atmosphere

【摘要】 微胶囊,是由具有一定厚度的密封壳层及其包裹的空间组成的一种结构,在药物的封装和缓释、催化等领域有着广泛的应用。微胶囊的制备方法有多种,其中一种常用方法是利用金属离子与配体间的配位相互作用制备的。本文以富勒醇为原料,通过向含有模板的富勒醇的酸性溶液中插入生锈的铁钉的方法,首次成功制备了富勒醇/铁微胶囊。详细研究了铁钉插入时间、富勒醇浓度和溶液酸度对微胶囊形成的影响,确定了铁钉插入2mg×m L-1富勒醇酸溶液(ph=2~4)中反应2 h为最终制备条件。研究了胶囊于不同的pH溶液中的分散稳定性,发现其在很宽泛的pH范围中(4~10)分散几天(?3)仍然能保留完整结构。所制备的微胶囊表现出铁磁性和良好的生物相容性。更有趣的是,电子顺磁共振测试表明微胶囊具有优良的羟基自由基猝灭性能,能有效提高细胞在H2O2气氛中的耐受性。

【Abstract】 Microcapsule, a supramolecular architecture consisting of a sealed shell with a certain thickness and an encapsulated space, has been widely used in drug encapsulation and release, catalysis and other fields. There are many methods to prepare microcapsules, one of which is by the coordination interaction between metal ions and ligands. Here, fullerenol/iron microcapsules were prepared successfully for the first time by inserting a rusted nail into a template-containing acidic fullerenol aqueous solution. The effects of insertion time of rusted iron nail, concentration of fullerenol and acidity of solution on capsule formation were studied in detail, and the final preparation condition was fixed as inserting rusty nails into 2 mg·m L-1 fullerenol acid solution(pH = 2~4) for 2 h. When the capsules were dispersed in aqueous solutions with varying pH for several days, not only a relatively wide range of pH stability, but also complete structures were observed. More interestingly, the fullerenol/iron capsules behaved ferromagnetic property, good biocompatibility and excellent OH×-quenching property, and could effectively improve the tolerance of cells in H2O2 atmosphere.
