碳纳米粒子对DPPC/DPPG单层膜成膜的实时影响

【摘要】 近年来空气污染问题日益严峻,在众多空气污染物中,PM2.5是人们最为关注的一种。PM2.5颗粒由于其体积小,在空气中悬浮时间长,可以进入肺部从而造成呼吸系统相关疾病,但是关于PM2.5颗粒物质通过呼吸进入肺部,以及其对于人体健康影响的具体动态影响机制方面的研究较为缺乏。本文利用Langmuir膜技术制备得到DPPC/DPPG单层膜,模拟人体肺部表面活性剂在呼吸过程中的成膜情况;在不同表面压的条件下加入碳纳米颗粒模拟人体呼吸过程中碳纳米颗粒从外界进入到肺部的动态过程;通过脂质单层膜的表面压力-平均分子面积(π-A)等温线、压缩模量-表面压力(-π)等温线、布儒斯特角显微镜(BAM)等探究了人体呼吸过程中碳纳米颗粒对肺部PS单层膜的实时影响。该工作不仅为深入理解呼吸过程中碳纳米颗粒对肺部的影响提供了详细信息,也为监测PM2.5对于人体肺部影响提供了新的研究思路。

【Abstract】 The air pollution is becoming more and more serious in recent years. As the most well-known air pollutant, fine particulate matter(PM2.5), has the small size and a longer suspension time. It also leads to some common respiratory diseases with inhaled into the lung to a certain concentration. There is a lack of research on the process of dynamically monitoring the real-time deposition of nanoparticles onto the PS monolayer. In this study, the DPPC/DPPG monolayer, prepared by the Langmuir-Blodgett method simulates the lung surfactant monolayers during respiration; the addition of carbon nanoparticles under different surface pressures are selected to simulate the real dynamic process of the human body inhales carbon nanoparticles during normal breathing. In this work, surface pressure(π)-area(A) isotherms of lipid monolayer, compressibility modulus(CS-1)-surface pressure(π) isotherms and the Brewster angle microscopy(BAM) were used to study the effect of carbon nanoparticles on the behavior of DPPC/DPPG monolayer in real-time. This study not only provides detailed information for in-depth understanding of the effects of carbon nanoparticles on lungs during respiration, but also opens up a new idea for real-time monitoring of the effects of PM2.5 to lung health.
