具有高效率双光子吸收,红光及近红外发射的AIEgen用于细胞器特异性成像以及成像辅助治疗

【摘要】 聚集诱导放光(AIE)材料因为其光稳定性强,生物兼容性好,发射波段可调,光动力效率高等特点,正在被开发用于细胞成像,光敏治疗等。本工作设计了一系列基于咔唑的具有强推拉电子效果的AIEgen,这些材料展现出高效率的双光子吸收,红光及近红外发射,强溶剂化效应,高效的固态发光等特点。这些材料具有良好的生物兼容性,通过结构的设计可以分别靶向到脂滴和线粒体两种细胞器,并且通过组织成像证明其具有较深的穿透。同时,在白光的照射下,这些材料可以高效的产生活性氧,用来杀伤癌细胞。这些材料具有很强的用于成像以及成像辅助治疗的潜力。
【Abstract】 Two-photon absorbing far red/near-infrared(NIR) fluorescent molecules with aggregation-induced emission(AIE) characteristics are much sought after for bioimaging and therapeutic applications in terms of imaging penetration depth and resolution especially if the fluorophores can be endowed with specific targeting capability. In this work, a carbazole-bridged dipolar far red/NIR-emissive AIE luminogens(AIEgens) with strong push-pull strength are synthesized and characterized. The obtained fluorophores display significant solvatochromism and TICT effect, bright far red/NIR solid state fluoirescence, large Stokes shifts, and large two-photon absorption cross-sections. They can target lipid droplet or mitochondrion specifically with tuned structure, and show deep penetrating depth in tissue imaging. At same time, they produce ROS very efficiently under white light and tailored into cancer ablation in vitro. These AIEgens hold great potential for cancer imaging and imaging-guided therapy.
