单细胞热敏通道的激活用于乳腺癌的治疗
Activation of single-cell thermosensitive ion channels for the treatment of breast cancer

【摘要】 纳米技术的医疗应用通常关注药物输送和生物传感器。使用外部光敏纳米材料控制细胞功能对于细胞工程技术和难治性疾病疗法的发展具有巨大的潜力。红外刺激为单细胞的电刺激提供了一种替代方案,具有在较高的空间分辨率下直接,非接触激活细胞的潜力。传统的INS方法受光在组织中穿透深度的限制。因此,在细胞表面负载抗体修饰的金纳米棒,采用780nm的NIR刺激表达TRPV1细胞。当局部温度升高时,TRPV1通道打开,大量钙离子内流,并在线粒体中聚集,使得钙激活的细胞凋亡途径打开,从而达到癌症治疗的目的。使用全细胞膜片钳电生理学观察TRPV1表达细胞的激光诱导离子通道的开放行为。通过用玻璃微电极测量细胞表面附近的瞬时温度的增加来确认由纳米棒引起的激光诱导加热。这些研究结果表明,这种健康的,可遥控的方法最终可能会应用于基础科学,技术和治疗。
【Abstract】 Medical applications of nanotechnology typically focus on drug delivery and biosensors. Controlling cell functions using external photoresponsive nanomaterials has enormous potential for the development of cell-engineering technologies and intractable disease therapies. Infrared stimulation offers an alternative to electrical stimulation of single-cells, with potential for direct, non-contact activation at high spatial resolution. Conventional methods of INS were limited by the depth of penetration of light in tissue. Therefore, instead with NIR at 780 nm to stimulate TRPV1-expressed cells, the surface of which were decorated with antibody-modified AuNRs. When local temperature rises, TRPV1 gates calcium to activation of apoptosis pathway through alterations in caspase activity. The laser induced ion channel opening behavior of TRPV1-expressed cell is observed using whole-cell patch clamp electrophysiology. The laser-induced heating by the nanorods is confirmed by measuring the transient temperature increase near the surface of the NR-ANs with an open pipette electrode. This healthy, remotely controllable method may eventually be applied to basic science, technology and therapy.
