基于超薄纳米片状配位聚合物的细胞内核酸检测与成像研究

【摘要】 二维(2D)纳米材料在分离、能源、催化、传感等方面逐渐引起关注。近年来,2D纳米材料广泛应用于细胞内核酸检测。发展低毒且高负载效率的核酸载体工具一直是研究的热点,而用于核酸转运的配位聚合物多为多孔材料,无孔的二维纳米片状配位聚合物在核酸转运方面还鲜有报道。本文基于无孔低维的Cu(I)三氮唑(tz)配位聚合物对单链DNA的强吸附作用,发展出一种高效、低毒的核酸运载工具进行细胞内核酸转运。Cutz纳米片一方面具有强的荧光淬灭性能,能淬灭探针链上的荧光;另一方面可以被癌细胞内的GSH及酸性条件降解,释放出被其淬灭的探针链的荧光。最终实现对单链DNA的负载,成功运载DNA进入细胞,其具有细胞内核酸的检测潜能。这种室温和水相条件便可简单制备的纳米片,有望代替Mn O2纳米片成为细胞内核酸(如microRNA)成像研究中的转运工具。

【Abstract】 Two-dimensional(2 D) nanomaterials have received increasing attention in separation, energy, catalysis, sensing, etc. Recently, 2 D nanomaterials have been employed widely in intracellular nucleic acid sensing. It is a hotspot to construct a low-cytotoxic and effective vehicle for intracellular delivery. While most of the coordination polymers(CPs) used in nucleic acid delivery are porous, the nonporous 2 D CPs were barely reported. Based on the strong adsorption of the nonporous 2 D Cu(I)-triazole(tz)-based CPs to single-stranded DNA, an efficient and low-cytotoxic tool was developed for intracellular nucleic acid transport. Cutz nanosheets have quenching ability towards fluorescently-labeled DNA, while they can be degraded by glutathione(GSH) and acidic conditions in cancer cells, resulting in the release of the probe’s fluorescence in cells. The nanosheets can be easily prepared at room temperature and aqueous conditions, and are expected to replace MnO2 nanosheets as a transport tool in intracellular nucleic acids(such as microRNA) sensing.
