基于低共熔溶剂的双重作用直接测定食用油中四种黄曲霉毒素

【摘要】 黄曲霉毒素是霉菌毒素中毒性最大,对人类健康危害最为突出的一类致癌物[1]。通常情况下,采用高效液相色谱-荧光技术对黄曲霉毒素测定时,都需要经过繁琐的提取、昂贵的亲和柱净化和衍生化处理[2],这样便增加了实验的复杂性和不稳定性[3]。基于此,本文以氯化胆碱为氢键受体,丙二酸为氢键供体加热制备了低共熔溶剂(DES),结合液相微萃取-液相色谱-荧光检测方法对食用油中四种黄曲霉毒素(G1、G2、B1和B2)进行分析。结果发现,DES对具有多羟基结构的G1、G2、B1和B2均具有良好的萃取与富集作用,并且经DES萃取后的G1和B1表现出很好的荧光性能。DES在液相微萃取过程中起到了萃取和衍生的双重作用,大大简化了油样前处理步骤,也显著降低了检测成本。具体实验过程和效果如图1所示。

【Abstract】 Aflatoxins, a kind of carcinogen, which are the most toxic and harmful to human health. At present, when HPLC-FLD is used to detect aflatoxins, it usually involves complex pre-processing, affinity purification and chemical derivatization, then the complexity and instability increase. In this paper, an one-step LMPE-HPLC-FLD method based on deep eutectic solvent(DES) was developed to analyse four aflatoxins from edible oils, in which DES was composed of choline chloride and malonic acid. We found that DES has a good effect on aflatoxins(G1, G2, B1 and B2), furthermore, G1 and B1 show fine fluorescence property after being extracted by DES. Thus, DES plays a dual role as extractant and derivatization agent in this work, which greatly simplifies oil sample pretreament and reduces the cost. The experimental process and chromatogramas are shown in figure 1.
