麦芽糖修饰磁性金属-有机骨架材料用于N-连接糖肽的高效富集

【摘要】 蛋白糖基化是最常见且重要的蛋白翻译后修饰之一,在分子识别、信号传导和免疫分析等生物进程中发挥重要作用[1]。因此,糖蛋白/糖肽的研究对临床疾病的早期诊断有重要意义[2]。本工作结合金属有机骨架和磁性材料的优点,利用简易的自组装策略,制备了一种具有核壳结构的麦芽糖修饰磁性金属有机骨架材料,作为亲水固定相富集N-连接糖肽。该材料具有亲水性好、比表面积大且麦芽糖固载量高的优点。在0.1pmol/uL HRP酶解产物中,该材料可以有效地富集24个糖肽。在200倍非糖肽干扰下,材料成功实现对糖肽的选择性富集。该方法简单易行且反应条件温和,有望将其应用于糖蛋白组学分析中低丰度糖肽的高效富集。

【Abstract】 glycosylation, as one of the most common and important post-translations modifications of proteins, as well as participates in many biological processes, such as molecular recognition, signal transdution and immune system response. Therefore, the specific profiling of glycopeptides and glycoproteins are significant for the early diagnosis of clinical diseases. In this work, combining the advantages of metal-organic framework and magnetic nanoparticles, a core-shell structure maltose-modified magnetic metal-organic framework was prepared via a facile self-assembly approach, and used as a novel hydrophilic material for enrichment of N-linked glycopeptides. Owing to its excellent hydrophilicity, large surface area and high immobilized amount of maltose, 24 glycopeptides were efficiently extracted from 0.1 pmol/uL digest of horseradish peroxidase(HRP). Besides, this nanocomposite show remarkable selectivity for N-linked glycopeptides even in the presence of 200 folds non-glycopeptides. The developed method is simple, mild reaction conditions and also provides a promising efficient and specific analytical platform in glycoproteome analysis.
