高效液相色谱-质谱联用技术鉴定长柄扁桃种皮多酚成分

【摘要】 长柄扁桃(Amygdalus pedunculata Pall),又名野扁桃、长梗扁桃,是蔷薇科扁桃属的落叶灌木。其种仁可用于提取优质食用油[1],但有关长柄扁桃种皮多酚组成及利用的研究尚未见报道。本实验采用微波辅助提取法提取了长柄扁桃种皮多酚,并用有机溶剂萃取法对多酚进行了纯化。运用高效液相色谱-电喷雾电离/质谱(LC-ESI/MS)对纯化后的多酚组成进行了分离检测,共分离鉴定出32种化合物,主要有芹菜素-7-O-葡萄糖苷、槲皮苷、山奈酚、芦丁、矢车菊素3-芸香糖苷、槲皮素、柚皮素和没食子酸等。其中芹菜素-7-O-葡萄糖苷的相对含量最高。这类物质普遍具有抗炎、抗氧化和抗癌活性[2],为后续长柄扁桃种皮多酚的生物活性研究提供依据。

【Abstract】 Amygdalus pedunculata Pall is a member of the family Rosaceae, and its seed is an excellent raw material for extracting high-quality edible oils. The polyphenolic constituents of Amygdalus pedunculata Pall seed coat(APSC) have not been reported. Microwave assisted extraction method was used to extract polyphenols from APSC, and the polyphenols were purified by organic solvent extraction. The composition of polyphenols was separated and detected by LC-ESI/MS. The 32 compounds were identified from APSC, main including apigenin-7-O-glucoside, quercitrin, kaempferol, rutin, cyanidin 3-rutinoside, quercetin, naringenin, and gallic acid et al. The highest content of the compound was apigenin 7-O-glucoside. These substances generally have anti-inflammatory, anti-oxidant and anti-cancer activities, which provide the basis for the subsequent research on the bioactive of the polyphenols in APSC.
