CD44和叶酸双受体靶向型荧光碳点的制备及细胞成像研究

【摘要】 由于荧光探针检测的高灵敏度、高特异性、高效快速等特性,双受体靶向荧光探针的研究策略开始应用于癌细胞的诊断[1]。但以荧光碳点为载体构建双受体靶向型荧光碳点的报道甚少,且碳点的发光性能较弱,迫切需要发展一种高灵敏度、发光性能良好的双受体靶向型荧光碳点。本文以D-(+)-氨基葡萄糖盐酸盐和L-天门冬氨酸为碳前驱体,通过一步水热法合成荧光性能优异的黄绿色荧光碳点。基于癌细胞过度表达的叶酸受体(FR)与CD44受体;利用叶酸与叶酸受体、透明质酸与CD44受体之间的特异性亲和作用,将叶酸与透明质酸依次以酰胺键枝接在聚乙烯亚胺(PEI)之上,再以碳点表面羧基与PEI残余氨基共价键合,制备了双受体靶向的荧光碳点,合成过程如图1(左)所示。图1(右)表明,合成的靶向碳点能够有效识别肿瘤细胞(A-D列)和正常细胞(E列),且不使用靶向分子掩蔽处理的肿瘤细胞(A列)展示出更强的荧光强度,说明合成的靶向碳点可用于双受体介导的肿瘤细胞精准荧光成像诊断。
【Abstract】 Spurred by the high sensitivity, specificity and efficiency in fluorescent probe, dual-targeted probes are quickly emerging as a strategy of choice for diagnosis of cancer cells. However, the complex reaction process and low fluorescence stability restrict the further application of fluorescent probes. One attractive strategy to solve this problem is developing dual-receptor targeted fluorescent CDs with high selectivity, sensitivity and good luminescence properties. In this research, yellow-green fluorescent carbon dots(CDs) coupled with hyaluronic acid and folic acid via amido bonds have been engineered for dual-receptor targeted cancer cells, due to the high specific affinity with hyaluronic acid, folate acid for CD44 receptors and folate receptors, respectively. The results show the strategy of combining CDs with hyaluronic acid and folic acid dual-targeted molecular will offer new potential directions in accurate fluorescence imaging diagnosis of tumor cells.
