空间分辨型电化学发光适配体传感器构建及癌胚抗原灵敏检测

【摘要】 构建了一种空间分辨的ECL适配体双检测模式传感平台,用于蛋白质的灵敏检测。CdS-C纳米复合材料和luminol-gold nanoparticles(L-Au NPs)作为该传感器ECL的阴极发光材料和阳极发光材料并修饰在双盘电极上,分别为工作电极1和工作电极2。然后固定上Au NPs、癌胚抗原(CEA)适配体的互补链(cDNA)和BSA后,用Ag-PAMAM NCs标记的生物素适配体(Ag-PAMAM-bio-Apt)与修饰过的双盘电极一起孵育。在这种情况下,由于Ag-PAMAM NCs在该ECL体系中对共反应剂H2O2的原位消耗,所以工作电极1和工作电极2上的ECL信号都会被猝灭。另外,L-Au NPs与Ag-PAMAM NCs之间存在ECL-RET,这会使工作电极2上的ECL强度进一步降低。在CEA存在下,由于CEA和cDNA之间对Ag-PAMAM-bio-Apt存在竞争反应而与双盘电极分离,从而增强了阴极和阳极的ECL强度。该方法可以实现对CEA的双重检测。阴极法的检测限为0.39 pg mL-1,阳极法的检测限为0.20pg mL-1。并将该方法应用于人血清中CEA的测定,回收率为80.0~110.0%,该方法为其他生物标志物的检测提供了一种新道路。

【Abstract】 A spatial-resolved ECL aptasensor for protein detection was constructed in a double-check mode.CdS-C as cathodic ECL emitters and luminol-gold nanoparticles (L-Au NPs) as anodic ECL emitters were immobilized on working electrode 1 (WE1) and working electrode 2 (WE2) of the dual-disk glassy carbon electrodes (DDCE),respectively.After successive immobilization of Au NPs,c DNA and BSA,the modified DDCE were incubated with Ag-PAMAM NCs-labeled biotin-aptamer (Ag-PAMAM-bio-Apt).Owing to the in-situ consumption of co-reactants H2O2 in this ECL system by Ag-PAMAM NCs,the ECL signals on both WE1 and WE2 were quenched.In addition,the ECL-RET existed between L-Au NPs and Ag-PAMAM NCs made the ECL on WE2 further decreasing.The presence of CEA generated an enhancement of ECL intensity on the DDCE.This strategy could achieve the detection of CEA in a double-check mode.The detection limits were 0.39 pg mL-1 for WE1 and 0.20 pg mL-1 for WE2,respectively.
