基于MOF的超灵敏双适配体电化学传感器用于特异性检测结核分枝杆菌

【摘要】 结核病是影响人类健康的重大疾病之一,但现有的检测方法存在耗时、成本高等缺点。MPT64是一种24-kDa蛋白,其特异性融合抗原在早期培养时仅分泌MTB复合物,可用于结核病的早期检测。我们选择MOF作为纳米粒子和酶的载体。制备了一种新的信号探针Au/UIO-66-NH2-HRP适配体。本文首次采用两个适配体的混合物检测MPT64,DPV结果表明它比使用单适配体效果更好。该电化学适配体传感器的线性范围为0.02-1000pg/mL,检出限低至10fg/mL。可用于早期检测结核分枝杆菌。
【Abstract】 School of Pharmaceutical Sciences, Sun Yat-Sen University, Guangzhou 510006, China Tuberculosis(TB)is a major health problem. There are many methods to detect TB which have some disadvantages. MPT64 can specifically fused antigen secreted only by MTB complex in early culture time. MOF is porous material which were chosen as the carrier of nanoparticles. A new signal probe Au/UiO-66-NH2-HRP-aptamer was fabricated. The mixture of two aptamers can specifically bind to MPT64 which have not been reported. The aptasensor performed well with good linear regression from 0.02-1000 pg/mL and the detection limit reached 10 fg/mL. The results demonstrate the mentioned method is specific and sensitive for the detection MPT64 in serum samples, proving the aptasensor to detect TB in early stage.
