槲皮素脂质体对糖尿病牙周炎大鼠牙周组织的作用

【摘要】 目的 :通过对Ⅱ型糖尿病牙周炎大鼠进行槲皮素干预,观察大鼠牙周组织的组织学变化以及血清AGEs的变化,探讨槲皮素对糖尿病牙周炎大鼠牙周组织的保护作用。方法:40只雄性SD大鼠,随机选取10只为空白对照组(N组),其余大鼠进行糖尿病牙周炎建模,将建模成功的20只大鼠随机均分为糖尿病牙周炎非治疗组(DP组)和糖尿病牙周炎槲皮素治疗组(DP+Q组),每组10只。DP+Q组灌喂槲皮素脂质体,DP组灌胃空白脂质体,给药6周后处死各组大鼠取上颌骨做牙周组织切片观察牙周组织病理变化,ELISA法测量血清AGEs含量。结果 :与DP组比较,DP+Q组大鼠牙周组织炎症水平明显较轻;DP组、DP+Q组AGEs含量高于N组,其中DP+Q组AGEs含量明显少于DP组(P<0.05)。结论 :槲皮素对糖尿病牙周炎大鼠的牙周组织有保护效果,其作用机制与降低血清AGEs含量有关。

【Abstract】 Objective: By establishing a rat model of type 2 diabetic periodontitis, observe the effect of quercetin on the periodontal tissue of experimental type 2 diabetic periodontitis rats and its effect on the level of advanced glycation end products(AGEs) in serum. Methods: 40 male SD rats, 10 were randomly selected as the blank control group(N group), and the remaining rats were modeled for diabetic periodontitis. The 20 rats that were successfully modeled were randomly divided into non-treatment groups for diabetic periodontitis( DP group) and diabetic periodontitis quercetin treatment group(DP+Q group), 10 rats in each group. The DP+Q group was fed with quercetin liposomes, and the DP group was fed with blank liposomes. After 6 weeks of administration, rats in each group were sacrificed. The maxilla was taken to make periodontal tissue slices to observe the pathological changes of periodontal tissues, which were measured by ELISA. Serum AGEs content. Results: Compared with the DP group, the level of inflammation in the periodontal tissue of the DP+Q group was significantly lighter; the levels of AGEs in the DP group and the DP+Q group were higher than those in the N group, and the AGEs content in the DP+Q group was significantly lower than that in the DP group(P <0.05). Conclusions: Quercetin has a protective effect on the periodontal tissues of diabetic periodontitis rats, and its mechanism is related to the reduction of serum AGEs content.
