乳酸促进口腔鳞癌干细胞自我更新的机制研究(英文)

【摘要】 <正>Purpose/Background:Accumulation of lactate within tumor is closely correlated with poor prognosis of patients with oral squamous cells carcinoma(OSCC), but the mechanism remains largely unknown. Presence of cancer stem cells(CSCs) contributes to initiation and progression of OSCC. The present study focus on role of lactate on self renewal of CSCs in OSCC.Materials and Methods:The primary OSCC cells were transfected with a Wnt reporter to visualize cells with CSCs properties. The CSCs population were enriched using sphere forming assay or flow cytometry based on Wnt activity level. The organoid reconstitution assay was employed for CSCs treated with lactate or lactate plus CHC, an inhibitor for MCT1. The oxygen consumption rate and ROS level were evaluated in OSCC-CSCs post lactate treatment.更多还原
