基于UPLC-QToFMS的骨关节炎血清代谢组研究

【摘要】 骨关节炎是临床常见的疼痛性和致残性关节炎疾病,其临床诊断尚缺乏有效的生物学标志物。本研究以31名骨关节炎患者和57名健康体检者为研究对象,运用基于超高效液相色谱-四极杆飞行时间串联质谱(UPLC-QToFMS)的非靶向代谢组学方法,检测其血清样本内源性代谢物质。借助有效的多维统计学和t检验数据分析方法,共筛查出46种骨关节炎相关的代谢物。研究结果表明,基于液质联用的血清代谢组学方法能够有效区分病例组和对照组,筛选出的潜在生物标志物有望用于骨关节炎的早期准确诊断和致病机制研究。
【Abstract】 Osteoarthritis(OA) is clinically the most common pain and disability arthritis.So far,reliable markers with high sensitivity and specificity for the accurate diagnosis of OA are still unavailable.In this work,based on untargeted metabolomics,ultra-performance liquid chromatography coupled with quadrupole time-of-fligit tandem mass spectrometry(UPLC-QToFMS)was performed to explore the serum metabolites from 31 OA patients and 57 healthy controls.Through the effective multivariate statistical analysis and t test analysis,46 differentially expressed metabolites were picked out and were regarded as OA-associated metabolites.Our results revealed that UPLC-QToF/MS-based metabolomics could effectively discriminate patients and healthy controls,and the selected potential biomarkers may be important for OA early diagnosis and pathogenic mechanism study in the future.
