SERS光谱检测单胺类氧化酶活性
SERS-Based Study on Enzyme Activity of Monoamine Oxidase

【摘要】 单胺类氧化酶是动物体内一类重要的含有黄素腺嘌呤二核苷酸(FAD)的线粒体外膜蛋白,可以降解儿茶酚、血清素等单胺类神经递质,其表达水平和活性的高低与一些纤维化疾病、抑郁症、老年精神疾病等有关。对氨基苯硫酚(PATP)作为探针分子在表面增强拉曼散射(SERS)光谱研究中被广泛地使用。MAO可以将单胺类底物氧化成醛,进而与PATP的氨基发生席夫碱反应。通过自组装方法得到了银(Ag)纳米粒子-PATP芯片,用以检测MAO催化产生的醛基产物,以达到体外甚至临床检测MAO酶活力水平的目的。
【Abstract】 Monoamine oxidase(MAO) is an important type of mitochondrial outer membrane protein containing flavin adenine dinucleotide(FAD) in animals,which can degrade monoamine neurotransmitters such as catechins and serotonin.The expression level and activity of MAO are related to some fibrosis diseases,depression and senile mental diseases.P-aminophenol(PATP) was widely used as a probe molecule in the study of surface enhanced Raman scattering(SERS) spectroscopy.MAO oxidizes monoamine substrates to aldehydes,which react with the amino group of PATP.We obtained a silver(Ag) nanoparticle-PATP chip by self-assembly method for the detection of MAO-catalyzed aldehyde group products, so as to achieve in vitro and even clinical detection of the level of MAO-enzyme activity.
