Pro-220对近红外荧光蛋白PAiRFP1光谱学行为的影响研究

【摘要】 以遗传学编码的光激活近红外荧光蛋白PAiRFP1为研究对象。运用基因工程以及光谱学等手段研究了Pro-220对蛋白光谱学行为的影响,结果发现当该位点Pro被丙氨酸取代后,突变体蛋白Pr态、Pfr态摩尔消光系数降低,近红外荧光量子产率也呈现下降趋势。上述结果表明:Pro-220对于PAiRFP1蛋白的近红外荧光发射发挥了重要作用,相关机理尚待深入研究。
【Abstract】 We hypothesize that the effects of spectroscopic behavior of proteins by using site directed mutation residues Pro220 A on genetically e1 coded light-activated near infrared fluorescent protein(PAiRFP1).Combining genetic engineering and spectroscopy results showed that when the pro line at this site was replaced by alanine,the molar extinctio, coefficient of the mutant proteins Pr and Pfr states decreased,and the quantum yield of near-infrared fluorescence also showed a downward trend.This result showed that Pro-220 play an important role in the near-infrared fluorescence emission of PAiRFP1 protein,and the related mechanism remains to be further studied.
