HOXA10表达时相对电针调控COH小鼠种植窗期的影响

【摘要】 目的:探讨电针调控COH小鼠种植窗期的机制。方法:240只雌性鼠和100只雄性鼠,选取40只雌性鼠设为自然周期组,剩余200只雌性鼠制备COH模型后,随机分为COH组、电针组、电针+阴性对照组、电针+增强HOXA10组和电针+抑制HOXA10组,每组40只。电针组、电针+阴性对照组、电针+增强HOXA10组和电针+抑制HOXA10取"关元"、"中极"、"三阴交"行电针治疗,自然周期组和COH组小鼠不予干预。将各组妊娠天数分为4小组,见阴栓第2d为孕2d,取孕6d得小鼠子宫,观察各组临床妊娠率和胚胎着床位点数,取孕3d、孕4d和孕5d小鼠子宫,采用Western Blot检测子宫内膜HOXA10蛋白表达。结果:电针干预及配合DNA脂质体复合物(增强HOXA10、抑制HOXA10)可改善COH组的妊娠率(P<0.05),在孕4d,电针治疗后,电针组、电针+阴性对照组、电针+增强HOXA10组的HOXA10蛋白较COH组明显增加(均P<0.05),且与自然周期组表达接近。结论:在针刺配合体外受精-胚胎移植(IVF-ET)治疗不孕症的过程中,HOXA10的表达影响到小鼠种植窗期的开放时间,针刺可通过调控COH小鼠种植窗期HOXA10的表达,从而提高临床妊娠率,改善胚胎着床。

【Abstract】 Objective To investigate the mechanism of electroacupuncture in regulating the window period of COH mice. Methods 240 female mice and 100 male mice, 40 female mice were selected as the natural cycle group, and the remaining 200 female mice were randomly divided into COH group, electroacupuncture group, electroacupuncture + negative control group, electroacupuncture + enhanced HOXA10 group and electroacupuncture +inhibited HOXA10 group, with 40 mice in each group. Electroacupuncture was performed in the electroacupuncture group, electroacupuncture + negative control group, electroacupuncture +enhanced HOXA10 group and electroacupuncture + inhibited HOXA10. Electroacupuncture(EA)was applied to "Guanyuan"(CV4), "Zhongji"(CV3) and "Sanyinjiao"(SP6) at the time points accordingly. EA stimulation was in the conditition of continuous wave, 2 Hz and 1 mA. No intervention was given in the natural period group and the COH group.The number of days of gestation in each group was divided into 4 groups. When the milky white solid "vaginal plug" was seen at the vaginal opening in female mice, we treated this as the first day(1 d) of gestation.Uterine tissues were collected at 3 d, 4 d, 5 d, and 6 d. The clinical pregnancy rate and embryo imbed site number were observed on 6 d. The uterus of mice with gestation 3 d, 4 d and 5 d was taken, and the expression of HOXA10 protein in endometrium was detected by Western Blot.Results Electroacupuncture intervention and DNA liposome complex(enhancing HOXA10 and inhibiting HOXA10) could improve the pregnancy rate of COH group(P < 0.05), After the treatment of 4 d pregnancy, the HOXA10 protein in the electroacupuncture group, the electroacupuncture + negative control group, and the electroacupuncture + enhanced HOXA10 group significantly increased compared with the COH group(all P < 0.05), which was close to the expression of the natural period group.Conclusion In the treatment of infertility by acupuncture combined with in vitro fertilization-embryo transfer(IVF-ET), the expression of HOXA10 affects the opening time of the implantation window of mice. Acupuncture can regulate the expression of HOXA10 in the implantation window of COH mice, thereby improving clinical pregnancy rate and embryo implantation.
