舒胃汤对功能性消化不良(FD)模型大鼠小肠推进功能及CNP/cGMP信号通路的影响

【摘要】 目的通过研究舒胃汤对功能性消化不良(FD)模型大鼠小肠推进功能及CNP/cGMP信号通路的影响,探讨舒胃汤治疗FD的作用机制。方法:取80只Wistar大鼠,按体质量随机分为空白组、模型组、舒胃汤组(3.068 g-kg-1)、莫沙必利组(1.37 mg·kg-1)。除空白组外其余组动物采用改良复合病因方法 (慢性束缚应激+过度疲劳+饮食失节+夹尾激怒)复制FD模型,造模结束后,连续灌胃给药2周。末次给药2 h后,脱颈处死大鼠,摘取胃窦环形肌条多道生理信号采集系统计算C-钠尿肽(CNP,1×10-8mol·L-1)对平滑肌张力的抑制率,WesternBlot法检测各组大鼠胃窦组织B型钠尿肽受体(NPR-B)蛋白含量,RT-PCR法检NPR-B mRNA表达,酶联免疫吸附法(ELISA)检测各组大鼠胃窦组织中环磷酸鸟苷(cGMP)含量。结果多道生理信号采集结果显示CNP对舒胃汤组大鼠胃窦环形平滑肌张力的抑制率明显降低(P<0.01)小肠推进实验结果显示舒胃汤组大鼠小肠推进速率明显升高(P<0.01);ELISA结果显示舒胃汤组大鼠cGMP蛋白含量明显降低(P<0.05) Western Blot结果显示舒胃汤组大鼠NPR-B蛋白表达明显降低(P<0.05);RT-PCR结果显示NPR-B mRNA表达明显降低(P<0.05)。结论:舒胃汤可有效改善FD模型大鼠胃肠功能低下状态,其机制可能与下调CNP/cGMP信号通路中CNP受体蛋白NPR-B表达以及关键蛋白cGMP含量有关。

【Abstract】 Objective:To explore the mechanism of Shuwei Decoction in the treatment of functional dyspepsia(FD) by studying the effect on Shuwei Decoction on intestinal propulsion and CNP/cGMP signaling pathway in rats with FD.Methods:80 Wistar rats were randomly divided into blank group,model group,Shuwei Decoction group(3.068 g-kg-1) and Mosapride group(1.27 mg-kg-1)Except the blank group,the other group of animal were treated with improved composite method(chronic restraint stress+excess fatigue+improper diet+xx) to create FD model.After the modeling,the rats were given continuous intragastric administration for 2 weeks.2 h after the last administration,rats were sacrificed,the gastric antral circular muscle strips were removed,the inhibitory rate of C-natriuretic peptide(CNP,1×10-8 mol/L) on smooth muscle tension was calculated by multichannel physiological signal acquisition system,B type natriuretic peptide receptor(NPR-B)protein in gastric antrum of rats was detected by Western Blot method,NPR-B mRNA expression was detected by RT-PCR method,and cyclonoside(cGMP) content was detected by enzyme linked immunosorbent assay(ELISA).Results:The results of multichannel physiological signal acquisition showed that the inhibitory rate of CNP on the tension of the circular smooth muscle of the gastric antrum in the Shuwei Decoction group was significantly lower(P<0.01);The experimental results of small intestinal propulsion showed that the small intestine propelling rate of the rats in Shuwei Decoction group was significantly increased(P<0.01);The results of ELISA showed that the content of cGMP protein in the rats of the Shuwei Decoction group was decreased(P<0.05);The results of Western Blot showed that the expression of NPR-B protein in the rats in Shuwei Decoction group was decreased(P<0.05);RT-PCR results showed that the expression of NPR-B mRNA was reduced(P<0.05).Conclusion:Shuwei Decoction can effectively improve the low gastrointestinal function of FD model rats,and the mechanism may be related to the downregulation of the expression of CNP receptor protein NPR-B and the content of key protein cGMP in the CNP/cGMP signaling pathway.
