用高内涵分析技术探究绒毛诃子肝毒性物质基础

【摘要】 目的:用高内涵分析技术(High Content Screening,HCS)研究绒毛诃子致肝毒性的物质基础。方法:采用染液对肝细胞进行标记后,利用ToxInsightIVT平台进行图像扫描,并且分析各个图像中各个指标的荧光强度。将各个至表皮分析所得的数据导入到DILI Assay Template中,从而得出受试药对细胞数目、DNA含量、GSH降低水平、ROS含量、MMP等指标的量效关系曲线,进而预测受试物是否有肝毒性。结果:阿司匹林和莽草酸结果为阴性,噻氯吡啶,苯甲酸,1,2,3,4,6-o-五没食子葡萄糖,没食子酸结果为阳性。结论:没食子酸肝毒性最大,没食子酸在给药浓度为50ug/ml时,较为安全。HCS在肝毒性预测中显示出的高灵敏度和特异性,作为一种快速、高效、高通量、多靶点的细胞毒性检测方法应广泛应用。
【Abstract】 Objective:To study the material basis of hepatotoxicity induced by HCS.Methods:Hepatocytes were labeled with dye solution,and images were scanned using the ToxInsight IVT platform,and the fluorescence intensity of each index in each image was analyzed.The data obtained from each epidermal analysis is imported into the DILI Assay Template to obtain a dose-effect relationship curve of the test drug on the number of cells,DNA content,GSH reduction level,ROS content,MMP,etc.,thereby predicting whether the test substance is Has liver toxicity.Results:Aspirin and shikimic acid were negative,and ticlopidine,benzoic acid,1,2,3,4,6-o-pentagalglucose gallic acid was positive.Conclusion:Gallic acid has the highest hepatotoxicity,and gallic acid is safer when the concentration is 50 ug/ml.HCS has shown high sensitivity and specificity in hepatotoxicity prediction,and it should be widely used as a rapid,efficient,high-throughput,multi-target cytotoxicity detection method.
