亚麻籽在慢性不可预知性应激大鼠中的抗抑郁作用
Antidepressant-like effect of flax seed in rats exposed to chronic unpredictable stress

【摘要】 目的:探究亚麻籽油和亚麻籽粕在慢性不可预知性应激大鼠中的抗抑郁作用。方法:50只大鼠随机分为5组:正常对照组(C,N=10,不接受任何实验性处理)、慢性不可预知性应激加生理盐水组(S-SN,N=10)、慢性不可预知性应激加艾司西酞普兰(S-Esc,N=10)、慢性不可预知性应激加亚麻籽油组(S-FO,N=10)和慢性不可预知性应激加亚麻籽粕组(S-FF,N=10);S-SN组、S-Esc组、S-FO组和S-FF组接受6周的慢性不可预知性应激,且在应激的第三周至第六周S-Esc组大鼠灌胃艾司西酞普兰(10 mg/kg/天),S-SN组大鼠灌胃等体积的生理盐水,S-FO组大鼠的饲料添加10%亚麻油,S-FF组大鼠的饲料添加20%亚麻籽粕。采用糖水偏爱测验和强迫游泳测验检测五组大鼠的抑郁水平,采用酶联免疫反应(ELISA)检测血清中脑源性神经营养因子(BDNF)浓度;采用免疫印迹法和实时荧光定量PCR(Real time PCR)检测海马内BDNF蛋白及BDNF mRNA的表达水平。结果:在糖水偏爱测验中,S-FO和S-FF组大鼠的糖水偏爱率显著高于S-SN和S-Esc组(p<0.05),且S-FO组显著高于S-FF组(p<0.05),S-Esc组显著低于C组(p<0.05);在强迫游泳测验中,S-FO组、S-FF组和S-Esc组大鼠的静止漂浮时间均显著短于S-SN组(p<0.05);S-FO组、S-FF组和S-Esc组大鼠的血浆BDNF浓度显著低于C组(p <0.01),且S-FO、S-FF组和S-SN组显著低于S-Esc组(p<0.05);在海马中,S-Esc组大鼠的BDNF蛋白浓度显著高于S-SN组(p<0.05),S-FO组大鼠的BDNF mRNA表达量显著低于C组(p<0.05)。糖水偏爱率与海马BDNF mRNA和蛋白表达量显著正相关(r=0.38,p<0.05;r=0.36,p<0.05);静止漂浮时间与血浆BDNF浓度、海马BDNF m RNA和蛋白表达量显著负相关(r=-0.44,p<0.05;r=0.32,p<0.05;r=0.29,p<0.05)。结论:亚麻籽油和亚麻籽粕在慢性不可预知性应激大鼠中抗抑郁作用,亚麻籽在抑郁症中可能有潜在的治疗作用。
【Abstract】 Objective: To explore the antidepressant-like effect of flaxseed oil and flour in rats induced by chronic unpredictable stress(CUS). Methods: Fifty male Sprague–Dawley(SD) rats were randomly divided into five groups. The first group was the normal control(C, N=10; no experimental treatment), the remaining four groups were exposed to CUS but forced fed with either: saline(S-SN, N=10; 10 ml/kg bodyweight per day); escitalopram(S-Esc, N=10; a standard antidepressant, 10 mg/kg bodyweight per day via lavage); flaxseed oil group(S-FO, N=10; feed containing 10% flaxseed oil), flaxseed flour group(S-FF, N=10; feed containing 20% flaxseed flour). Behaviors were tested using sucrose preference test and forced swimming test. The serum BDNF concentration, hippocampal BDNF m RNA and protein expression were measured by ELISA, real-time PCR, and Western blot, respectively. Results: The sucrose preference rate was significantly higher in S-FO and S-FF rats than S-SN and S-Esc rats(p < 0.01), higher in S-FO rats than S-FF rats(p < 0.05), and lower in S-Esc rats than C rats(p < 0.01). The immobility time was significantly shorter in S-FO, S-FF rats and S-Esc than S-SN rats(p < 0.05). Plasma BDNF concentrations were significantly lower in S-FO, S-FF, and S-Esc rats than C rats(p < 0.01), lower in S-FO rats, S-FF and S-SN than S-Esc rats(p < 0.05). The hippocampal BDNF protein expression was significantly higher in S-Esc rats than S-SN rats(p < 0.05). The hippocampal BDNF mRNA expression was significantly lower in S-FO rats than C rats(p < 0.05). The sucrose preference rate was positively correlated with hippocampal BDNF mRNA and protein expression(r=0.38,p<0.05;r=0.36,p<0.05). While the immobility time was negatively correlated with plasma BDNF concentration, hippocampal BDNF mRNAand protein expression(r=-0.44,p<0.05;r=0.32,p<0.05;r=0.29,p<0.05) Conclusion: Flaxseed oil and flaxseed flour exert antidepressant-like effect in rats exposed to chronic stress. flaxseed may have a therapeutic effect on depression.
