酶促合成DHA-PC中脂肪酸组成的气/液相色谱分析方法比较

【摘要】 研究证明,二十二碳六烯酸-磷脂(DHA-PLs)比DHA-乙酯、DHA-甘油三酯和游离DHA氧化稳定性更好、生物利用率更高、营养功能更全面。课题组以天然大豆磷脂酰胆碱(PC)为原料,在南极假丝酵母脂肪酶B的催化作用下,使其与藻油发生酰基转移反应制得DHA-PC。本文建立了以α-溴苯乙酮为衍生化剂,乙腈/甲醇/水为梯度洗脱剂,在波长244 nm,柱温50℃下进行的柱前衍生化-高效液相色谱法检测DHA-PC中脂肪酸的组成,并与传统的气相色谱分析法作了比较。试验结果表明:用柱前衍生-液相色谱法检测DHA-PC中的脂肪酸,各组分可以完全分离且峰型良好,其主要成分的批内精密度(n=6)和稳定性(n=6)的相对标准偏差(RSD)分别为2.5%~3.2%和1.9%~4.0%,平均回收率为97.8%~106%。测定结果与气相色谱法有很好的一致性。由此可知,柱前衍生-高效液相法可以克服气相色谱检测温度高可能引起不饱和脂肪酸异构化的不足,用于稳定、准确的测定DHA-PC中脂肪酸的组成。
【Abstract】 Compared with docosahexaenoic acid(DHA)-ethyl ester,DHA-triglycerides and free DHA,DHA-phospholipids(DHA-PLs) have attracted more attention due to their advantages in oxidation stability,bioavailability efficiency and nutritional functions.DHA-PC was synthesized by CalB-mediated transesterification of phosphatidylcholine(PC) and algal oil containing triglyceride of DHA(DHA-TG).An efficient pre-column derivatization high-performance liquid chromatography(HPLC) analysis method was proposed to measure the fatty acid concentration of DHA-PC.The mobile phase was acetonitrile-methanol-water for gradient elution at 244 nm and a column temperature of 50 ℃,then compared with the traditional gas chromatography(GC).Results show that the intra-assay precisions(n=6) and stabilities(n=6) relative standard deviations(RSDs) for the major components were 2.5%-3.2% and 1.9%-4.0%,and the average recoveries were97.8%-106% for pre-column derivatization HPLC.In addition,the good agreement with the results from GC means that the lower detection temperature may avoid the isomerization of unsaturated fatty acids in DHA-PC.In summary,it indicated that pre-column derivatization HPLC.was rapid,accurate and reliable to analyze the content of DHA-PC in the phospholipid product.
