小麦TaGaBp基因特征及其在干热胁迫下的表达分析

【摘要】 TaGaBp是小麦中最重要的Ca2+感受器之一,能够与Ca2+结合来响应外界干热环境,该家族蛋白参与小麦对环境的适应过程。本研究借助生物信息学对小麦TaGaBp基因进行分析,结果表明TaGaBp基因位于小麦染色体4B上,具有6个外显子,CDS序列长度为1206 bp,蛋白序列长度为402个氨基酸,pI为5.26。进化分析表明小麦TaGaBp与大麦TuGaBp关系最近。启动子顺式作用元件分析表明TaGaBp基因启动子区域存在CGTCA-motif,G-Box,GC-motif,GT1-motif,LAMP-element,MBSI,LTR,MRE,O2-site,TC-rich repeats,ABRE,ABRE3a,ABRE4等与生长发育、激素信号传导及逆境胁迫响应相关的顺式作用元件。RT-PCR结果表明干热胁迫能增加苗期小麦根组织中TaGaBp的表达量,说明TaGaBp参与小麦对干热胁迫的响应,对小麦TaGaBp基因功能的进一步研究将为小麦耐热育种改良提供理论依据。
【Abstract】 TaGaBp is one of the most important Ca2+receptors in wheat.It can bind with Ca2+to respond to dry and heat environment.TaGaBp family proteins are involved in the adaptation process of wheat to the environment In this study,TaGaBp gene in wheat was analyzed by bioinformatics.The results showed that TaGaBp gene was located on chromosome 4 B of wheat with six exons.The length of CDs sequence was 1206 bp,the length of protein sequence was 402 amino acids,and PI was5.26.Phylogenetic analysis showedthat wheat TaGaBp had the closest relationship with TuGaBp.Promoter cis acting elements analysis showed that CGTCA-motif,G-Box,GC-motif,GT1-motif,LAMP-element,MBSI,LTR,MRE,O2-site,TC-rich repeats,ABRE,ABRE3 a,ABRE4 and other cis acting elements related to growth and development,hormone signal transduction and stress response were found in the promoter region of TaGaBp gene.The results of RT-PCR showed that dry heat stress could increase the expression of TaGaBp in root tissue of wheat at seedling stage,which indicated that TaGaBp was involved in the response of wheat under dry heat stress.Further study on the function of wheat TaGaBp gene will provide theoretical basis for the improvement of wheat heat tolerance breeding.
