基于MXene@ LDH异质结构的高灵敏谷胱甘肽检测
MXene@LDH heterostructures for sensitive colorimetric detection of glutathione

【摘要】 谷胱甘肽(GSH)是一种重要的含硫醇三肽,广泛存在于哺乳动物和真核细胞中,在人体代谢过程中也起着自由基信号转导,基因调控和对异生物的细胞抗性等作用。研究表明,一些疾病的发生(例如,肝损害、白细胞降低和癌症等)往往与细胞内GSH的异常变化有直接关系;然而,GSH的检测往往依赖于复杂的仪器设备、繁琐的预处理以及较高的检测成本,并且当前的检测技术在选择性、抗干扰和稳定性等方面仍无法满足临床和医学需求。本研究制备了一种新型的2D/2D异质结构,由MXene纳米片和层状双氢氧化物纳米片组成的MXene@LDH催化剂,并首次将其用于谷胱甘肽的检测研究。MXene具有优异的金属导电性、高亲水性和良好的机械性能,为催化反应中电子的传递提供了快速通道,同时为LDH提供了良好载体,避免了LDH的团聚。通过有效结合MXene和LDH以及界面工程,制备出一种优异的纳米酶材料。将该MXene@LDH用于比色法谷胱甘肽检测,获得了优异的检测灵敏性、选择性和稳定性,并且在人体血清实验中也获得了较好的检测结果。
【Abstract】 Glutathione(GSH) is an important thiol-containing tripeptide that widely exists in mammalian and eukaryotic cells.It serves essential functions in many metabolic processes,such as free radical signal transduction,gene regulation,and cellular resistance against xenobiotics.The occurrence of some disease(e.g.liver damage,leucocytes loss,and even cancers) is often directly linked to the abnormal level of GSH in our human cells.However,the current GSH detection technologies normally require the use of sophisticated instruments,tedious pretreatment,or high detection cost;plus,the detection performances cannot meet the clinical and medical requirements in terms of their poor selectivity,anti-interference and stability.In our work,a 2 D/2 D heterogeneous structure,consisting of MXene supported layered double hydroxide nanoflakes(denoted as MXene@LDH),was synthesized and for the first time is applied for the GSH detection.MXene was a new group of 2 D nanomaterials that characteristics of metallic conductivity,hydrophilicity,and good mechanical properties.It not only provided fast electron transfer channels during catalytic reactions,but also reduced the agglomeration of LDH.The synergistic effect of two phases and the interface engineering effect made MXene@LDH being an excellent nanoenzyme.As a result,the MXene@LDH exhibited a superior detection sensitivity,selectivity and stability for the colorimetric glutathione detection.The established sensor was also applied in the detection of GSH from human serum and displayed satisfying results.
